【摘要】 目的 观察螺内酯对T2DM大鼠脂肪组织脂联素、PPARγmRNA表达的影响。 方法 高脂饲养加小剂量链脲佐菌素注射建立T2DM大鼠模型。雄性SD大鼠50只,分正常组(NC,n=10),糖尿病对照组(DM-C,n=15)和糖尿病干预组(DM-S,n=14),实验末留取血液和附睾脂肪组织,检测血液学指标和脂肪组织脂联素、PPARγ的表达水平。 结果 DM-C组和DM-S组血TG均高于NC组(P<0.01), DM-S组水平低于DM-C组(P<0.05)。脂肪组织中脂联素和PPARγmRNA表达,DM-C组低于NC组[(4.26±0.5) vs (4.85±0.24),(5.12±0.28) vs (5.43±0.22),P<0.05],DM-S组高于DM-C组[(4.72±0.33 )vs (4.26±0.5),(5.36±0.22 )vs (5.12±0.28),P<0.05]。 结论 螺内酯上调T2DM大鼠脂肪组织中脂联素及PPARγmRNA的表达,降低血TG,这可能是改善糖尿病IR和靶器官损伤的一个潜在机制。

【Abstract】 Objective To investigate the effect of spironolactone on the expression of adiponectin and peroxisome proliferator activated receptor gama (PPARγ) mRNA in epididymal adipose tissue of type 2 diabetic rats. Methods The model of T2DM rats were established through high-sucrose-high-fat diet and followed by low dose Streptozotocin injecting. Fifty healthy male Sprague-Dawley rats were divided into 3 groups: Normal control rats (NC), diabetes control group (DM-C), and diabetic rats treated with Spironolactone (DM-S). The group DM-S was treated with Spironolactone via intragastric administration for 8 weeks. Serum markers were tested and the mRNA expression of adiponectin and PPARγin adipose tissue were detected. Results Triglyceride level was significantly higher in group DM-C and DM-S compared with group NC (P<0.01) and it was lower in group DM-S than in group DM-C (P<0.05). The mRNA levels of adiponection and PPARγ were lower in group DM-C than in group NC [(4.26±0.5) vs (4.85±0.24) and (5.12±0.28) vs (5.43±0.22), (P<0.05)], and were significantly increased in group DM-S as compared with group DM-C [(4.72±0.33) vs (4.26±0.5) and (5.36±0.22) vs (5.12±0.28), (P<0.05)]. Conclusion Spironolactone increases expression of adiponectin and PPARγ mRNA in adipose tissue and reduces serum TG level of type 2 diabetic rats, which may involve a potential mechanism for protection against insulin resistance and lessening tissue damage in diabetes.
