Exendin-4对游离脂肪酸介导的βTc6细胞GK和GLUT2表达改变的干预作用

  【摘要】 目的 探讨Exendin-4在高脂状态下是否具有抗脂毒性及其相关的机制是否涉及葡萄糖激酶(GK)和葡萄糖转运蛋白2(GLUT2)。方法 应用不同浓度Exendin-4(5、10及50nmol/L)孵育βTc6细胞不同时限(6、12及24h),再予1 mmol/L FFA作用24 h后,检测细胞内GK和GLUT2表达情况。结果 Exendin-4干预6 h,各组细胞内GK、GLUT2表达未见明显改善;当干预延长至12~24 h,随干预浓度增高,细胞内GK、GLUT2表达逐步升高。 结论 适宜浓度的Exendin-4干预一定的时限可上调脂毒性状态下胰岛β细胞GK和GLUT2的表达。

  【Abstract】 Objective To investigate the anti-lipotoxicity of Exendin-4 in lipotoxic state and whether its mechanism is related with glucokinase (GK) and glucose transporter 2 (GLUT2). Methods βTc6 cells were incubated with Exendin-4 (5, 10 and 50nmol/L) for different duration (6,12 and 24h) prior to intervention with 1mmol/L free fatty acids (FFA) for 24 hours, then GK and GLUT2 expression was examined. Results When treating with Exendin-4 for 6 hours, the expression of GK and GLUT2 in βTc6 cells was not up-regulated in each group. After treating with Exendin-4 for 12~24 hours, the expression of GK and GLUT2 was increased gradually along with increasing the concentration of Exendin-4. Conclusion Treating with Exendin-4 in optimal concentration and duration, the expression of GK and GLUT2 can be up-regulated in lipotoxic state.
