胰腺星状细胞在2型糖尿病模型大鼠胰岛纤维化进程中作用的观察

  【摘要】 目的 观察胰腺星状细胞(PSC)在T2DM模型GK大鼠胰岛中的存在状态,探讨PSC在胰岛纤维化发生发展进程中的作用。 方法 GK与Wistar大鼠胰腺冰冻切片行Masson染色和荧光双标(α-SMA/Insulin)染色。采用外向生长法分离纯化GK大鼠胰岛生细胞,免疫荧光检测Vimentin,α-平滑肌肌动蛋白(α-SMA),Fibronectin(FN)和Col I/III的表达。 结果 GK大鼠胰岛纤维化区域α-SMA染色阳性细胞增多,胰岛素分泌缺失。大量PSC自GK大鼠胰岛内生外生,表达Vimentin、α-SMA、FN和Col I/Ⅲ。 结论 活化PSC存在于T2DM模型GK大鼠纤维化胰岛中,并在T2DM胰岛纤维化进程中发挥重要作用。

  【Abstract】Objective To observe stellate cell (PSC) existing in GK rat islets and explore the role of PSC in the progression of islet fibrosis. Methods Masson staining and immunofluorescence staining assay (α-SMA/insulin) were performed using frozen sections of GK/Wistar rat pancreas. Isolating PSC by outgrowth method, and immunofluorescence staining assay for α-SMA, Vimentin, Fibronectin and Col type I / III were done for the observation. Results The α-SMA positive cells increased and beta cell mass decreased in the fibrotic area in GK rat islet. Large number of PSC grew out from GK rat islets, and these cells expressed α-SMA, Vimentin, FN and Col I/III. Conclusion The activated PSC presented in fibrotic islets of GK rats and they may play an important role in T2DM islet fibrosis.
