抵抗素299G/A和过氧化物酶体激增物激活受体γProl2Ala基因多态性与妊娠期糖尿病的相关性研究

  【摘要】 目的 探讨抵抗素299G/A、PPARγPro12Ala基因多态性与GDM的相关性。 方法 选取156例GDM孕妇为GDM组和180名正常对照(NC)孕妇为NC组,采用聚合酶链反应-限制性内切酶技术(PCR-RFLP)对基因多态性进行检测。 结果 GDM、NC组抵抗素299G/A基因型频率GG:GA:AA分别为41.3%:46.3%:11.6%、35.2%:51.7%:13.3%,PPARγProl2Ala基因型频率PP:PA:AA分别为92.3%:7.7%:0.0%、85.0%:13.3%:1.7%,两组间基因型和等位基因频率比较差异均无统计学意义。GDM组抵抗素基因 GA、AA基因型孕妇FIns、胰岛素抵抗指数(HOMA-IR)高于GG基因型[(12.99±9.98) vs (8.06±2.25)mU/L,(18.35±6.32) vs (8.06±2.25)mU/L;(2.45±2.32) vs (1.31±0.36),(3.45±1.89) vs (1.31±0.36),P<0.05]。PPARγ基因PP基因型孕妇BMI高于PA基因型[(29.47±1.68) vs (23.89±2.88)kg/m2,P<0.01]。 结论 抵抗素299G/A、PPARγPro12Ala基因多态性与GDM发病无相关性,可能与GDM患者肥胖、IR有关。

  【Abstract】Objective To investigate the correlation ofresistin299G/A, PPARγPro12Ala polymorphism with gestational diabetes mellitus(GDM). Methods Selecting156 cases of pregnant women with GDM as GDM group, and 180 cases of normal control (NC) pregnant women as NC group, polymerase chain reaction and restriction endonuclease technique (PCR-RFLP) were employed for genotypingresistin299G/A, PPARγProl2Ala polymorphisms. ResultsResistin299G/A polymorphism genotype frequencies of GG:GA:AA were 41.3%:46.3%:11.6% respectively for GDM group and 35.2%:51.7%:13.3% for NC group,.PPARγProl2Ala polymorphism genotype frequencies of PP:PA:AA were respectively 92.4%:7.6%:0.0% for GDM group and 85.0%:13.3%:1.7% for NC group. There was no difference between two groups of genotype and allele frequency. The levels of fasting insulin (Fins) and HOMA-insulin resistance (IR)in GDM groupwere higher inresistin GA and AA genotypes than inGG genotype [Fins(12.99±9.98)vs(8.06±2.25)mU/L, (18.35±6.32)vs(8.06±2.25)mU/L, respectively; IR(2.45±2.32)vs(1.31±0.36), (3.45±1.89)vs(1.31±0.36), respectively; all P<0.05]. BMI in GDM group was significantly higher in PP genotype than in PA genotype [(29.47±1.68)vs(23.89±2.88)kg/m2, P<0.01].ConclusionResistin299G/A and PPARγPro12Ala polymorphism may have no relationship with the incidence of GDM, but may have a certain relationship with obesity and IR.
