沉默信息调节因子对高糖诱导的系膜细胞内皮素-1和转化生长因子β1的关系研究

[摘要] 目的   探讨沉默信息调节因子(SIRT1)对高糖诱导的系膜细胞内皮素-1ET-1)和转化生长因子β1TGF-β1)的关系。 方法  体外培养系膜细胞分为:(1)高糖组(高糖培养液);2SIRT1激动剂(白藜芦醇)+高糖组,含1μmol/L白藜芦醇的高糖培养液;3SIRT1 RNAi干扰+高糖组,加入pTRC-shSIRT1慢病毒感染;4正常对照(NC)组。检测各组细胞SIRT1 基因表达,以及ET-1TGF-β1水平。 结果高糖组、SIRT1激动剂+高糖组、SIRT1 RNAi干扰+高糖组及NC ET-1含量分别为(56.1±6.3)、(31.9±5.1)、(105.63±8.2)和(17.3±3.4pg/mlTGF-β1含量分别为(43.3±5.7)、(27.5±4.9)、(87.4±7.3)和(15.6±3.6ng/ml。高糖抑制SIRT1表达,上调ET-1TGF-β1表达(P0.05);SIRT1激动剂可改善高糖对SIRT1的抑制,下调ET-1TGF-β1表达(P0.05);沉默SIRT1,可上调ET-1TGF-β1表达(P0.05)。  结论  SIRT1基因对高糖诱导的系膜细胞ET-1TGF-β1可能起负调控作用。

Relationship of silent information regulator (SIRT1)withendothelin-1 and transforming growth factor β1 in mesangial cells induced by high glucose

AbstractObjective To investigate the relationship ofSIRT1 withendothelin-1 (ET-1) and transforming growth factor β1 (TGF-β1) in mesangial cells induced by high glucose.    Methods Mesangial cells(RMC) cultured in vitro were obtained and divided into fourgroups: high glucose group (cultured inhigh glucose medium); SIRT1 agonist (resveratrol) + high glucose group (cultured inhigh glucose medium with1μmol/Lresveratrol;SIRT1 RNAi + high glucose group (transfected by pTRC-shSIRT1in high glucose medium); normal control group (NC group). Expression of SIRT1 gene weredetected. ET-1 and TGF-β1 were measuredby Elisa.Results ET-1leveland TGF-β1 level were56.1±6.3,31.9±5.1,105.63±8.2, 17.3±3.4pg/mland43.3±5.7,27.5±4.9,87.4±7.3, 15.6±3.6ng/mlin high glucose group, SIRT1 agonist (resveratrol) + high glucose group, SIRT1 RNAi + high glucose group and NC grouprespectively.Expression of SIRT1 gene was inhibited in high glucose environment.Expressions of ET-1 and TGF-β1were also up-regulatedin high glucose state (P < 0.05).The inhibition of high glucose on SIRT1 gene was improved and the expressions of ET-1 and TGF-β1 were down-regulatedby SIRT1 agonist(P < 0.05). The expressions of ET-1 and TGF-β1 were significantlyup-regulated by silencing SIRT1 gene (P < 0.05).     Conclusion    SIRT1 gene acts as a negative regulator forET-1 and TGF-β1 levels in mesangial cells induced by high glucose.
