姜黄素对糖尿病大鼠肾脏血管紧张素Ⅰ型受体表达的影响

【摘要】 目的  观察姜黄素对糖尿病大鼠肾脏血管紧张素Ⅰ型(AT1)受体表达的影响。方法  选取将瘦型及肥胖型Zucker大鼠各16只,随机分为瘦型溶剂(Lean Vehicle)组,肥胖型溶剂(Obese Vehicle)组,瘦型姜黄素组及肥胖型姜黄素组。分别给予溶剂或姜黄素(2.0 mmol/L)口服喂养4周后,ELISA法测定血清丙二醛(MDA)和谷胱甘肽(GSH)水平。qRT-PCRWestern 印迹法测定肾脏AT1受体mRNA及蛋白表达。 结果  肥胖大鼠氧化应激显著增强(MDA:6.1±0.6 vs 3.5±0.8; GSH: 3.4±0.5 vs 7.4±0.7 nmol/mL)(P<0.05)。肾脏AT1受体mRNA及蛋白表达也高于瘦大鼠(P<0.05)。姜黄素处理后,可明显降低肥胖大鼠氧化应激指标(MDA: 4.1±0.5GSH: 5.3±0.6, nmol/mLP<0.05),同时AT1受体mRNA及蛋白表达也降低(P<0.05结论  姜黄素可降低肥胖大鼠氧化应激及肾脏AT1受体的表达,促进其血压的降低。

Effect of curcumin on the AT1 receptor expression in the diabetic rat kidney XU Jian-hui, YANG Bo (Department of Cardiology, Peoples Hospital of Wuhan UniversityHubei, 430060

Abstract】  Objective  To investigate the effect of curcumin on the angiotensin receptor type-1 inkidney of obese rats.  Methods  Lean and obese rats in each group were used in this study. These rats were randomly divided into lean vehicle group, obese vehicle group, lean curcumin group and obese curcumin group. Control lean and obese rats received drinking water, Lean and obese rats were provided with curcumin (2.0 mmol/L in drinking water) for a duration of 4 wk. The levels of MDA and GSH in serum were measured by ELISA. The mRNA and protein expression levels of AT1 receptor were measured by qRT-PCR and Westernblotting, respectively. Results The levels of MDA were higher and the GSH lower in obese rats as compared with lean rats (MDA: 6.1±0.6 vs 3.5±0.8; GSH:3.4±0.5 vs 7.4±0.7, nmol/mL) (P<0.05). Moreover, the AT1 receptor mRNA and protein expressions were also higher in obese rats (P<0.05). Curcumin treatment for 4 weeks reduced oxidative stress in obese rats (MDA:4.1±0.5; GSH:5.3±0.6, nmol/mL)(P<0.05). Moreover, AT1 receptor mRNA and protein expression were reduced in obese rats (P<0.05). Conclusion curcumin reduces oxidative stress and expression of AT1 receptor in kidney of obese rats, which would be helpful to lower the blood pressure of obese rats.
