血管生成素2与血管内皮生长因子和2型糖尿病大血管病变的关系

   【摘要】 目的 探讨血管生成素2(Ang-2)与血管内皮生长因子(VEGF)与T2DM大血管病变的关系。方法 用ELISA法测定40例T2DM合并大血管病变(MVC)、30例T2DM无大血管病变(Non-MVC))及30例正常者对照组(NC)的Ang-2和VEGF的血清水平。结果 MVC组的Ang-2及VEGF血清水平明显高于Non-MVC组和NC组(Ang-2中位数为4.62ng/ml比2.85ng/ml和1.89ng/ml,P均<0.01;VEGF中位数为297.5pg/ml比181.4pg/ml和96.1pg/ml, P<0.05,<0.01),Non-MVC组高于NC组(P<0.05);Tie-2在三组间差异无显著性(P>0.05)。在MVC组,Ang-2与VEGF及空腹胰岛素呈正相关(r值分别为0.524 ,0.404,P<0.01、<0.05)logistic回归分析显示,GHbA1c、Ang-2、糖尿病病程、VEGF与T2DM大血管病变独立相关。 结论 血清Ang-2及VEGF表达水平与2型糖尿病大血管病变相关,Ang-2协同VEGF在2型糖尿病大血管发生和发展进展中可能起着重要作用。

  【Abstract】 Objective To study the possible association between angiopoietin 2(Ang-2), vascular endothelialium growth factor(VEGF) and the pathogenesis of macroangiopathy in type 2 diabetes(T2DM). Methods Serum Ang-2 and VEGF were measured by enzyme-linked immunosorbent assay, which were determined on 40 cases T2DM with Macrovascular Complications (MVC), 32 cases without macroangiopathy (Non-MVC), and 30 healthy control subjects(NC) .Results Serum levels(median) of Ang-2 and VEGF in MVC were significantly increased compared with Non-MVC and NC group (Ang-2: 4.62ng/ml vs 2.85ng/ml and 1.89ng/ml, P<0.01); VEGF: 297.5pg/ml vs. 181.4pg/ml and 96.1pg/ml, P<0.05,P<0.01, respectively); Ang-2 and VEGF were higer in Non-MVC than in NC group (P<0.05). As for Tie-2, No statistically significant differences were found among the three groups. In MVC, Spearman correlation analysis indicated that Ang-2 showed positive correlation with VEGF,Tie-2, and FIns levels((r=0.524, 0.404, P<0.01,P<0.05, respectively). Multivariate logistic regression analysis showed that GHbA1c、Ang-2、the duration of diabetes、VEGF were significantly associated with the development of macroangiopathy in T2DM. Conclusion The study suggests that Serum angiopoietin-2 and VEGF levels are associated with initiation and progression of T2DM macroangiopathy, and meanwhile Ang-2 together with VEGF possibly play an important role in the pathological course of Atherosclerosis.
