BDNF Val66Met与红水河流域壮族长寿家系血糖水平缺乏相关性

  【摘要】 目的 探讨红水河流域壮族长寿家系(HLF)BDNF Val66Met(rs6265)多态性与血糖水平的相关性。 方法 采用ARMS-PCR对HLF (n=1531)rs6265位点进行基因分型,并与当地非长寿家系(HNLF, n=600)及外地区(平果)长寿家系(PLF, n=662)和非长寿家系(PNLF, n=545)进行比较,分析基因型与血糖水平的关系及其在长寿中的作用。 结果 (1)HLF rs6265等位基因G和A频率分别为56.3%和43.7%,与其他家系的频率分布差异有统计学意义(P = 0.024)。总体上,平果地区A等位基因频率高于红水河流域,PLF中A的频率有低于PNLF的趋势,而HLF A频率则有高于HNLF的趋势;HLF基因型频率分别为GG 31.2%、GA 50.2%、AA 18.6%,与其他对照家系比较差异无统计学意义。(2)PLF GA/AA基因型携带者的血糖水平低于GG携带者(P = 0.038)。(3)多元回归分析未发现各家系中血糖水平与基因型的相关性。结论 BDNF Val66Met多态在红水河地区的分布有异于外地区,可能是该流域长寿的遗传学基础之一,但与血糖水平的关系不大。

No association of BDNF Val66Met polymorphism with blood glucose in Zhuang People long-lived families living along Guangxi Hongshuihe River Basin WU Jin, LV Ze-ping , PAN Shang-ling, et al. Department of Pathophysiology, Guangxi Medical University, Nanning 530021, China


  【Abstract】 Objectives To evaluate the putative correlation between BDNF Val66Met polymorphism (rs6265) and serum glucose in the longevity families living along the Basin of Guangxi Hongshuihe River. Methods BDNF Val66Met locus was genotyped using ARMS-PCR ,and genotyping results were compared among 4 groups of Guangxi Hongshuihe long-lived (HLF, n= ) and non-long-lived families(HNLF, n=), Guangxi Pingguo long-lived (PLF, n=662) and non-long-lived families (PNLF, n=545). The correlation and its role in longevity between BDNF Val66Met genotypes and serum glucose were analyzed. Results (1) The frequencies of G and A in rs6265 allele of HLF were 56.3% and 43.7, respectively. This allelic frequency distribution significantly differred from those of other three counterparts(P = 0.024), in which A allele was overall more highly expressed in Pingguo dwellers than Hongshuihe dwellers. In addition, A allele tended to be lower in PNLF as compared with PLF, while it seemed to enrich in HLF when compared with its geographic matched control (HNLF); the genotypic frequencies of rs6562 (GG, GA, AA) in HLF were 31.2%, 50.2% and 18.6%, respectively, which appeared similar to other controls.(2)A lower glucose level was noted in A allele (GA/AA) carriers than in non-A carriers (GG) in PLF.(3) No relationship between serum glucose and genotypes was observed in multiple regression analysis. Conclusion The polymorphism of BDNF Val66Met in Hongshuihe inhabitants differs from that of other area, which may be one of the molecular bases of its longevity. However, no association is found between the polymorphism and serum glucose level.
