【摘要】 目的 探讨盐酸小檗碱(BER)对胰岛β细胞内质网应激相关凋亡的影响。 方法 以衣霉素(Tun)诱导胰岛β细胞株MIN6细胞内质网应激,检测BER对细胞增殖和凋亡的影响,对胰岛素分泌的影响,及对细胞凋亡相关蛋白ASK1、Caspase-12表达的影响。 结果 BER(1 μmol/L、10 μmol/L)对内质网应激细胞基础和葡萄糖刺激的胰岛素分泌均无明显影响,可减少细胞凋亡、抑制凋亡相关蛋白ASK1和Caspase-12的表达。 结论 BER能减少胰岛β细胞内质网应激相关凋亡,其机制可能与抑制凋亡相关蛋白ASK1和Caspase-12的表达有关。

【Abstract】 Objective To explore the effect of berberine on the endoplasmic reticulum stress-related apoptosis in pancreatic β cells. Methods The endoplasmic reticulum stress in MIN6 cells was induced by Tunicamycin. The effect of berberine on cell proliferation and apoptosis, insulin secretion, expression of apoptosis-related proteins ASK1 and Caspase-12 were examined. Results Basal and glucose-stimulated insulin secretion were not affected while cell apoptosis was improved by berberine (1μmol/L, 10μmol/L), however, the expression of ASK1 and Caspase-12 was inhibited significantly by berberine. Conclusions Berberine can reduce the endoplasmic reticulum stress-related apoptosis in pancreatic β cells. The mechanism may be related to the inhibition of ASK1 and Caspase-12 expressions.
