【摘要】 目的 探讨中国汉族人群p38βMAPK基因多态性对mRNA表达的影响。 方法 纳入非糖尿病正常人群98名,经测序分析rs2072878位点AA基因型82人，AG基因型16人。分别选取AA基因型组与AG基因型组各14人，提取外周血单核细胞(PBMC)，提取RNA与蛋白。p38βMAPK mRNA的测定采用实时荧光定量PCR，p38 MAPK总蛋白表达的检测采用SDS-PAGE 电泳-蛋白免疫印迹法。 结果 p38βMAPK rs2072878的AA基因型人群 mRNA表达水平显著低于AG基因型人群[(0.485±1.143) vs (2.250±2.719)，P=0.034]。p38MAPK蛋白表达水平AA基因型组与AG基因型组差异无统计学意义[(1.347±0.309)vs(1.007±0.500)，P=0.375]。 结论 中国人群p38βMAPK rs2072878基因态性可能影响其mRNA表达，而对p38MAPK 总蛋白的表达没有影响。

　　【Abstract】Objective To examine the effect of genetic variations in the p38ß MAPK genes on its mRNA levels in the normal Hans screened by 75-g OGTT. Methods Totally 98 unrelated normal subjects were genotyped by Mass spectrometry assay, the carriers of AG were 16 and AA 82 respectively, and then 14 subjects were selected from each genotype group. The mRNAs and proteins from PBMC were extracted, the mRNA level of p38ß MAPK was detected by Real-time PCR (Taqman Probe), and the total protein level of p38 MAPK was estimated by SDS-PAGE western blotting analysis. Results The AA carriers at rs2072878 in p38ß MAPK gene had lower mRNA levels compared with the AG carriers [(0.485±1.143) vs (2.250±2.719), p=0.034]. There was no significant difference in total protein levels of p38 MAPK between the two genotype groups [(1.347±0.309) vs (1.007±0.500), p=0.375]. Conclusions Common variant(s) in p38ß MAPK gene may impact its mRNA expression in normal Han population in Beijing, while it may have no effect on the protein expression of p38 MAPK .
