腺苷酸活化蛋白激酶对糖尿病大鼠非酒精性脂肪性肝病的影响

  【摘要】 目的 观察腺苷酸活化蛋白激酶(AMPK)在糖尿病大鼠肝脏的表达,探讨其在非酒精性脂肪肝病(NAFLD)发病中的作用。方法 40只雄性SD大鼠随机分为正常对照(NC)组、饮食诱导肥胖(DIO)组、糖尿病(DM)组。采用Western-blot方法测定肝脏AMPK蛋白表达;HE染色后观察肝脏形态学变化。结果 DIO组血清FIns、HOMA-IR及肝脏TG含量均较NC组升高;DM组FIns、HOMA-IR较NC组及DIO组升高;DIO组肝AMPK蛋白较NC组降低(P<0.05),DM组肝AMPK蛋白表达较DIO组降低(P<0.05),DM组肝脏磷酸化腺苷酸活化蛋白激酶(p-AMPK)表达较NC组降低(P<0.01)。结论 AMPK可能参与了肥胖及糖尿病大鼠NAFLD的发生发展。

  【Abstract】 Objective To observe the expression level of AMPK in liver of fatty and diabetic rats by a high fat diet,and to investigate the role of AMPK in the pathogenesis of NAFLD. Methods Forty male Sprague-Dawley rats were randomly divided into normal control group, diet-induced obesity (DIO) group and diabetes mellitus group. The protein expression of AMPK was measured by Western blot. Also the morphology change of liver was observed by light microscopy stained with HE. Results Liver TG, FIns, HOMA-IR were significantly higher in DIO rats than in NC group(P<0.05). FIns and HOMA-IR were significantly higher in DM group than in group NC and DIO(P<0.05). The protein expression of AMPK of DIO rats were significantly lower than that of group NC(P<0.05).The protein expression of AMPK were significantly lower in DM rats than group DIO(P<0.05). The protein expression of p-AMPK were significantly lower in DM rats than in group NC(P<0.01).Conclusion AMPK probably takes part in the pathogenesis of nonalcoholic fatty liver disease in obesity and diabetic rats .
