PPARγ2基因Prol2Ala多态性对2型糖尿病 相关指标的影响

  【摘要】 目的 从基因水平探讨过氧化物酶体增殖物激活受体γ2(PPARγ2)基因Pro12Ala多态性对T2DM患者有关指标的影响。 方法 选取广东省汉族T2DM患者328例,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术对PPARγ2基因Pro12Ala多态性的基因型进行检测,并测定相关指标。 结果 与 Pro12 Pro组比较,Pro12Ala 组T2DM患者TC、LDL-C、瘦素(LP)水平和胰岛素抵抗指数(HOMA-IR)差异有统计学意义(P<0.05)。将患者按肥胖组(BMI>25 kg/m2 ,170例) 和非肥胖组(BMI≤25 kg/m2,158例)分层后,肥胖组TC、TG、LDL-C的水平Pro12Ala组高于Pro12 Pro组,差异有统计学意义(P<0.05)。 结论 PPARγ2基因Pro12Ala多态性可能影响T2DM患者TC、LDL-C、LP及HOMA-IR。

  【Abstract】 Objective To investigate the influence of peroxisome proliferation activated receptorγ2 (PPARγ2) gene Pro12Ala polymorphism on the serum lipid of type 2 diabetes mellitus (T2DM) in Chinese people. Methods The 328 T2DM patients of Han nationality of Guangdong province were selected for investigation. The polymorphism of PPARγ gene Pro12Ala was detected by PCR-RFLP, and its relevant indicators were determined as well. Results Compared with the Pro12 Pro group, the levels of total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), leptin (LP) and Homeostasis model assessment of insulin resistance (HOMA-IR) in the Pro12Ala group were different significantly (P<0.05). The patients were divided into obese group (BMI>25kg/m2, n=170) and non-obese group (BMI≤25kg/m2, n=158). The levels of TC, TG, and LDL-C of obese Pro12Ala group were higher than those in the Pro12 Pro group, and the difference was significant in statistics (P<0.05). Conclusion PPAR-γ2 gene Pro12Ala polymorphism may affect the levels of TC,LDL-C,LP, and HOMA-IR of the T2DM patients.
