  α2β1整合素Bgl II基因多态性与2型糖尿病肾病相关性分析

  【摘要】 目的 研究α2β1整合素基因多态性与糖尿病肾病(DN)之间的相关性。 方法 采用PCR-RFLPs法,对298例南京区T2DM患者(合并肾病182例,无肾病116例)和107例正常人进行α2β1 BglII基因多态性研究。 结果 ⑴ DN组Bg1Ⅱ+/+基因型高于无肾病组(DM组)和正常对照(NC组)(P<0.01),与NC组比,DN组患者Bg1Ⅱ+等位基因频率增加(P<0.05);⑵ T2DM患者Bg1Ⅱ+/+型血清肌酐、尿微量白蛋白排泄率高于Bg1Ⅱ+/-及Bg1Ⅱ-/-型(P<0.05);Bg1Ⅱ基因型各亚组间胰岛素抵抗指数、β细胞功能指数无统计学差异(P>0.05);(3) Bg1Ⅱ基因型各亚组间血脂水平无显著差异(P>0.05)。 结论 α2β1整合素BglII基因多态性与DN存在相关性,Bg1Ⅱ+等位基因可能是DN的危险因子之一。

  【ABSTRACT】Objective To investigate the association between α2β1 integrin Bgl Ⅱ gene polymorphism and diabetic nephropathy (DN). Methods In this study, there were 298 T2DM patients involved, of them, 182 were accompanied with DN and 116 without DN, and 107 healthy people were selected to be the normal controls (NC). PCR-RFLPs technique was adopted for detecting the α2β1 integrin Bg1Ⅱ gene polymorphism. Results (1) BglⅡ+/+ genotype frequency of the DN patients was higher than those without DN and NC group (P<0.01). Compare with the NC group, Bg1Ⅱ+ allele frequency of DN patients combined with DN was increased (P<0.05). (2) Serum creatinine and urinary albumin excretion rate of T2DM patients with Bg1Ⅱ+/+ genotype were higher than those of the patients with Bg1Ⅱ+/- and Bg1Ⅱ-/- genotypes (P<0.05). The insulin resistance index and β-cell function index of T2DM patients with different Bg1Ⅱ genotypes were not different statistically (P>0.05). (3) Plasma lipid levels of the T2DM patients with different Bg1Ⅱgenotypes were not significantly different (P>0.05). Conclusion The α2β1 integrin Bg1Ⅱ gene polymorphism of is associated with DN, and the Bg1Ⅱ+ allele polymorphism may be a risk factor for DN of the T2DM patients.
