血浆miR-126在1型糖尿病患者中的变化及其临床意义

     【摘要】 目的   探讨T1DM患者血浆miR-126表达水平的变化及其临床意义. 方法   选取T1DM患者36例及健康对照(NGT)者34名,RT-PCR检测各组血浆miR-126,并分析其与年龄、BMI、高敏C反应蛋白(hsC-RP)、内皮型一氧化氮合酶(eNOS)、FPG、2 hPG、HbA1C、FC-P、2 hC-P、HDL-C、LDL-C、TG、TC的关系。 结果   与NGT组比较,TIDM组血浆miR-126表达下降[(1.65±0.61)VS
(0.68±0.18),P<0.053。相关分析显示,miR-126与HDL-C、FC-P、2 hC-P及eNOS呈正相关(P<0.05或P<0.01),与LDL-C、TC、hsC-RP及年龄呈负相关(P<0.05或P<0.01);多元逐步回归分析显示,hsC-RP、FC-P、TC、年龄是miR-126的影响因素. 结论   TIDM患者血浆miR-126呈低表达,可能与胰岛功能及内皮功能有关,hsC-RP及TC是miR-126的独立影响因素,miR-126可能参与了TIDM患者的炎症反应及脂质代谢异常,且miR-126水平随年龄增长而下降。

    【Abstract】 Objective   To investigate the changes of plasma miR-126 expression leveI in patientswith type 1 diabetes(T1DM). Methods   The plasma miR-126 expression level was investigated in 36T1DM patients and 34 healthy controls (NGT)by real-time fluorescent quantitative polymerase chainreaction(PCR).The relationship between plasma miR-126 and clinical indicators such as age,BMI,hsC-RP,eNOS,FPG,2 hPG,HbA1c, FC-P, 2 hC-P,HDL-C,LDL-C,TG and TC was analyzed by correlation
analysis.The main influencing factors for miR-126 were analyzed by multiple stepwise regression analysis.Results   The miR-126 expression level was significantly decreased in T1DM group than that in NGT group[(1.65±0.61)VS(0.68±0.18),P<0.05].Correlation analysis showed that miR-126 was positivelycorrelated with HDL-C,FCP,2 hC-P and eNOS(P<0.05 or P<0.01),and negatively correlated with LDL-C,TC,hsC-RP and age(P<0.05 or P<0.01).Multiple stepwise regression analysis showed that hsC-RP,FC-P,TC and age were independent influencing factors for miR-126.    Conclusion    The expression level of plasma miR-126 is lower in TIDM group than in NC group,which is possibly associated with islet and endothelial functions.hsC-RP and TC are independent influencing factors for miR-126,which may be involved in the inflammatory reaction and 1ipid metabolism disorder in T1DM patients.With age,the expression level of miR-1 26 may decrease.
