血清炎症因子在OLETF鼠2型糖尿病发病过程中的动态变化

   【摘要】 目的 探讨自发性T2DM大鼠 模型OLETF鼠发病过程中炎症因子与IR的关系。 方法 20只雄性OLETF鼠(OLETF组)和10只雄性LETO鼠(LETO组)在8、32和40周龄分批处死。检测血糖和血浆胰岛素水平。免疫散射比浊法检测各组大鼠血清超敏C反应蛋白(hsC-RP),ELISA法检测血清肿瘤坏死因子α(TNF-α)、白介素6(IL-6)、单核细胞趋化蛋白1(MCP-1)水平,并计算ISI。 结果 8周龄时,OLETF组血清hsC-RP和MCP-1高于LETO组;随周龄增加,OLETF组血清hsC-RP、TNF-α、IL-6、MCP-1水平逐渐升高,且40周龄时,均高于LETO组,仅ISI低于LETO组(P<0.01)。32周龄和40周龄时,ISI与hsC-RP、IL-6呈负相关(P<0.01),血清各炎症因子之间互为正相关(P<0.05)。 结论 OLETF鼠血清炎症因子在糖尿病发生前即升高,并随病情进展持续升高,与IR密切相关,参与T2DM的发生发展。

  【Abstract】 Objective To analyze the dynamic expression of serum inflammatory markers during the development of diabetes in OLETF rats. Methods 20 male OLETF rats (OLETF group) and 10 male LETO (LETO group) rats were executed separately at age of 8, 32 and 40 weeks. The fasting blood glucose and plasm insulin were determined. Serum hsC-RP was determined by the immunonephelometry assay. Serum TNF-α, IL-6, MCP-1 were measured by Enzyme-linked immunoadsordent assay (ELISA) and calculated insulin sensitivity index (ISI) . Results The serum hsC-RP and MCP-1 in OLETF rats were significantly higher than LETO rats at 8-week age. The levels of serum hsC-RP、TNF-α、IL-6、MCP-1 in OLETF group were increased with week age ,and were higher than LETO group at 40-weeks age,but ISI was lower(P<0.01). ISI was negatively correlated with hsC-RP and IL-6 at rats of 32 and 40 weeks of age(P<0.01). All the inflammatory markers were positively correlated(P<0.05). Conclusions Serum inflammatory markers increase in OLETF rats before the onset of type 2 diabetes mellitus, and along with the state continue to rise,which are closely correlated with insulin resistance and contribute to the onset and development of type 2 diabetes mellitus.
