实验性低血糖对糖尿病大鼠血清ET-1的影响

  【摘要】 目的 探讨实验性低血糖和血糖波动对DM大鼠ET-1的影响。方法 正常Wistar大鼠随机分为对照组(NC)和低血糖组(NL),DM大鼠随机分为对照组(DC)、单纯低血糖组(DL)和低血糖后高血糖组(DLH)。低血糖大鼠用胰岛素诱导低血糖。DLH组于低血糖后给予50%葡萄糖灌胃诱发高血糖。所有实验大鼠于胰岛素或生理盐水注射前、注射后1.5h、4.5h、6.5h取血测定BG和血清ET-1。 结果 低血糖各组血清ET-1水平明显升高(DLH>DL>NL);NL组ET-1水平在BG最低点时最高(116.83±11.68);DL组和DLH组ET-1水平在BG最低点时均无升高,峰值呈延迟现象;DLH组BG再度升高时ET-1水平开始升高,峰值显著高于DL组(239.39±22.22 vs 161.12±55.94,P<0.05)。 结论 低血糖对DM大鼠血管内皮损伤更明显;血糖剧烈波动对血管内皮损伤比持续低血糖更严重。

  【Absrtact】 Objective To ascertain the influence of insulin-induced acute hypoglycemia and glucose fluctuation on the serum endothelin 1 (ET-1) of diabetic mice. Methods Healthy male Wistar mice were randomized into control group (NC) and hypoglycemia group (NL). Diabetic male Wistar mice were randomized into control group (DC), hypoglycemia group (DL), and hypoglycemia followed by hyperglycemia group (DLH). Hypoglycemia was induced by intraperitoneal administration of insulin. Hyperglycemia was induced in the DLH group by intragastric administration of 50% glucose solution. Blood samples were obtained at the time before injection and 1.5h, 4.5h, and 6.5h after injection to detect blood glucose and ET-1. Results The level of ET-1 in each hypoglycemic group increased significantly (DLH>DL>NL). It reached the maximum when the blood glucose in the NL group was to the minimum (116.83±11.68). It didn’t increase in the DL and DLH groups when the blood glucose was to the minimum, with a delayed presence of its peak. It began to increase when the blood glucose was higher again in the DLH group, with a significantly higher peak than in the DL group (239.393±22.22 vs 161.12±55.94 P<0.05). Conclusion Hypoglycemia causes more serious injury of endothelium cells of diabetic mice, and glucose fluctuation induces more serious injury of endothelium cells than uncontrolled hypoglycemia.
