糖尿病动脉粥样硬化大鼠血清中二羰基化合物含量的观察及其临床意义

  【摘要】 目的  观察糖尿病动脉粥样硬化(AS)大鼠血清中二羰基化合物的表达。方法:20只2型糖尿病Goto-Kakizaki (GK)大鼠采用左旋硝基精氨酸甲酯(L-NAME)和高脂饲料喂养法制备糖尿病动脉粥样硬化大鼠模型,另选10只Wistar大鼠作为对照组。应用气相色谱-质谱联用仪(GC-MS)对大鼠血清进行分离、纯化,测定其中二羰基化合物甲基乙二醛(methylglyoxal,MG)和3-脱氧葡萄糖醛酮(3-Deoxyglucosone,3-DG)的含量。结果:糖尿病动脉粥样硬化模型组大鼠血清中二羰基化合物MG和3-DG含量分别为216.7±15.6μmol/L和19.5±3.8μmol/L,与对照组(85.3±24.9μmol/L和8.2±2.4μmol/L)相比,含量明显升高(P<0.05)。结论:二羰基化合物含量升高可能是糖尿病动脉粥样硬化的发病机制之一。

  【Abstract】 Objective To detect the concentration of dicarbonyl compounds in serum of diabetic atherosclerosis rats. Methods Twenty Goto-Kakizaki (GK) rats were randomized into the control and model groups. The diabetic atherosclerosis rat models were produced by feeding with NG-Nitro-L-arginine methyl ester (L-NAME) and high fat and sucrose diet. Another 10 Wistar rats were taken as the controls. Gas chromatography-mass spectrometry (GC-MS) was applied to determine the concentration of dicarbonyl compounds methylglyoxal (MG) and 3-Deoxyglucosone (3-DG) in serum.

  Results The concentration of MG and 3-DG in diabetic atherosclerosis rats (216.7±15.6μmol/L and 19.5±3.8μmol/L respectively) was significantly higher than in the control group (85.3±24.9μmol/L and 8.2±2.4μmol/L respectively) (P<0.05). Conclusion The elevated levels of dicarbonyl compounds may be one of the mechanisms of diabetic atherosclerosis.
