α-硫辛酸对糖尿病大鼠下丘脑和骨骼肌组织腺苷酸活化蛋白激酶表达及活性的影响

  α-硫辛酸对糖尿病大鼠下丘脑和骨骼肌组织腺苷酸活化蛋白激酶表达及活性的影响

  【摘要】 目的 通过观察α-硫辛酸对糖尿病大鼠腺苷酸活化蛋白激酶(AMPK)的影响,探讨其改善糖尿病代谢紊乱的作用。方法 将大鼠分为3组:正常对照(NC)组、糖尿病对照(T2DM)组、糖尿病α-硫辛酸干预(LA)组。Western blot检测下丘脑及骨骼肌组织AMPKα及ph-AMPKα蛋白的表达,RT-PCR测AMPKα mRNA表达。结果 与NC组比较,T2DM组骨骼肌及下丘脑内AMPK表达水平(21%和22%)及活性(37%和21%)降低(P<0.05),血糖、血脂水平升高;与T2DM组比较,LA组大鼠骨骼肌内AMPK表达水平(20%)及活性(49%)升高(P<0.05),而下丘脑内AMPK表达水平(17%)及活性(36%)降低(P<0.05),血糖、血脂水平降低。结论a-硫辛酸可激活骨骼肌AMPK,抑制下丘脑AMPK活性,可能是其改善糖脂代谢紊乱的部分机制。

  【Abstract】 Objective To observe the effect of α-lipoic acid on the adenosine monophosphate-activated protein kinase (AMPK) in the diabetic rats and to research the function of AMPK in improving the metabolism turbulence. Methods Wistar rats were randomized into 3 groups: normal control group (NC group), T2DM control group (T2DM group), and diabetes α-lipoic acid intervention group (LA group). The diabetic models were established with STZ. Western blot assay was adopted to detect the expression of AMPKα and ph-AMPKα, and RT-PCR to observe the expression of AMPKαmRNA. Results Compared with the NC group, the level of expression (21% and 22%) and activity (37% and 21%) of AMPK were decreased in the hypothalamus and skeletal muscle of the T2DM group (P<0.05), while the level of blood glucose and lipid was increased. Compared with the T2DM group, the expression (20%) and activity (49%) of AMPK in the skeletal muscle of the LA rats were increased (P<0.05), while the expression (17%) and activity (36%) of AMPK in the hypothalamus were decreased (P<0.05), also the level of blood glucose and lipid was decreased. Conclusion a-lipoic acid can activate the AMPK in the skeletal muscle and suppress it in the hypothalamus of diabetic rats. This may be a partial mechanism of a-lipoic acid improving the metabolism turbulence.
