PPAR-γ激动剂吡格列酮对大鼠骨髓内皮祖细胞凋亡的抑制作用及其机制探讨

  PPAR-γ激动剂吡格列酮对大鼠骨髓内皮祖细胞凋亡的抑制作用及其机制探讨

  【摘要】目的 探讨选择性过氧化物酶体增殖物激活受体γ激动剂吡格列酮(PIO)对大鼠骨髓内皮祖细胞(EPCs)凋亡的影响及其机制。方法 使用密度梯度离心法及差速贴壁法联合的方法培养大鼠骨髓EPCs,并使用双荧光染色法进行鉴定。将大鼠骨髓EPCs置于含不同质量浓度PIO培养基中,检测EPCs凋亡情况。EPCs培养7d后分为5组,分别加入一定浓度的PIO、PPARγ拮抗剂GW9662、PIO及 PI3K/Akt通道阻滞剂(Wortmannin)、PIO及ERK通道阻滞剂PD98059,检测EPCs的凋亡情况。 结果 PIO为50μmol/L时,可最大程度抑制EPCs的凋亡,凋亡率为(2.39±0.37)%。这种作用在PI3K/Akt通道阻滞剂Wortmannin及PPAR-γ拮抗剂GW9662存在时作用消失,而在ERK通道阻滞剂PD98059作用下仍存在。结论 PIO可抑制EPCs的凋亡,该作用可能是通过PI3K/Akt信号通路介导。

  【Abstract】 Objective TO explore the effect of selective peroxisome proliferator-activated receptor γ agonists pioglitazone (PIO) on the apoptosis of endothelial progenitor cells’ (EPCs) and to study its functional mechanism. Methods EPCs were cultured by using density gradient centrifugation and differential velocity adherent methods, and the double dyeing method was used to identify. Culturing EPCs in medium containing pioglitazone of different concentrations, and setting up a blank control group for comparison, the EPCs’ apoptosis was determined. After culturing for 7 days, EPCs were divided into 5 groups, and then pioglitazone, pioglitazone and PPAR antagonists GW9662, pioglitazone and PI3K/Akt channel blocker Wortmannin, pioglitazone and ERK channel blocker PD98059 were added respectively for observing the EPCs’ apoptosis. Results When pioglitazone at 50 μmol/L concentration , it could inhibit EPCs apoptosis at the greatest degree, the apoptosis rate was (2.39±0.37)%. This effect disappeared when there were PI3K/Akt channel blocker Wortmannin and PPAR-γ antagonist GW9662 in action, however this effect was still existing under the action of ERK channel blocker PD98059. Conclusion Pioglitazone can inhibit EPC apoptosis. This effect may be mediated by PI3K/Akt signal pathway.
