  β细胞功能缺失后的分子学和细胞学机制研究新领域:Id1基因—β细胞功能的新型抑制剂

  [提要] Id1属于四种Id蛋白质(Id1—4)的家族,是螺旋-环-螺旋(HLH)家族转录因子的成员。Id蛋白被称为“DNA结合抑制剂”。目前,T2DM胰岛β细胞功能障碍的有关分子机制仍未明确,但在糖尿病小鼠的胰岛中以及β细胞功能障碍的体外模型中已经发现HLH蛋白Id1的表达有所增加。为了考察了Id1对胰岛素分泌和葡萄糖稳态的作用,Mia C. Åkerfeldt和D. Ross Laybutt进行了相关研究,结果发现,Id1缺失小鼠高脂饲料喂养后未发生糖尿病,且体内和体外β细胞功能增强。现将相关研究和评论的精华部分编译如下,与广大读者共享。

  A new frontier for the research of molecular and cellular mechanisms behind the lack of properβ-cell function: Id1 — a novel inhibitor of β-cell function HONG Shan-shan, QIAN Rong-li. Editorial Department of Chinese Journal of Diabetes, Beijing 100034, China

  【Summary】Id proteins are named “inhibitor of DNA binding”. The molecular mechanisms responsible for pancreatic β-cell dysfunction in T2DM remain unresolved. Increased expression of the helix-loop-helix (HLH) protein Id1 has been found in islets of diabetic mice and in vitro models ofβ-cell dysfunction. Mia C. Åkerfeldt and D. Ross Laybutt investigated the role of Id1 in insulin secretion and glucose homeostasis, and found that Id1-deficient mice were protected against diabetes following high-fat feeding, and that β-cell function in vivo and in vitro was enhanced in these mice devoid of Id1.Here we translate essential parts of the related article and commentary and presented them in the following.
