糖尿病神经性疼痛大鼠脊髓背角星形胶质细胞的变化

  【摘要】 目的 观察糖尿病神经性疼痛(DNP)大鼠脊髓星形胶质细胞的变化。方法 SD大鼠30只,分为正常对照(NC)组10只和糖尿病(DM)组20只。DM组腹腔单次注射STZ(80 mg/kg),NC组腹腔注射生理盐水。2周后眼眶内眦取血测定血糖值,血糖>16.7 mmol/L大鼠作为糖尿病模型。4周后测定大鼠热板反应潜伏时间作为痛阈指标,选取DM组痛阈最低的10只作为DNP模型组。取DNP组大鼠脊髓腰段组织切片。神经胶质纤维酸性蛋白(GFAP)标记星形胶质细胞,观察星形胶质细胞在脊髓背角的分布;采用多媒体彩色病理图像分析系统检测阳性面积(Area)和灰度值(Gray Scale)。 结果 DM组痛阈(18.41±6.25)较NC组(37.40±7.35)明显降低(P<0.01),DNP组GFAP染色阳性脊髓星形胶质细胞面积(1377.37±271.83)和灰度值(8638.21±1861.90)水平较NC组明显升高(P<0.01)。 结论 糖尿病神经性疼痛时大鼠脊髓星形胶质细胞被激活增生,表达水平升高,抑制脊髓星形胶质细胞激活可能成为治疗DNP的新策略。

  Response of astrocyte in the spinal cord of rats with diabetes neuropathic pain

  【Abstract】Objective To observe the response of astrocyte in the spinal cord of rats with diabetes neuropathic pain. Methods Thirty SD rats were randomized into normal control (NC) group (n=10) and diabetes (DM) group (n=210). Diabetes was induced by peritoneal injection with streptozotocin (STZ, 80mg/kg). The NC group rats were administered 1ml saline. Two weeks later the blood was obtained from the inner canthus and the level of blood glucose was measured. Those rats with the blood glucose >16.7mmol/L were of the diabetes models. Hot plate latent time was recorded as pain threshold in the fourth week. Diabetes neuropathic pain (DNP) model was confirmed when pain threshold decreased significantly. The rats were sacrificed and their spinal cords were taken at the end of the 4th week. The distribution of astrocytes marked by glial fibrillary acidic protein (GFAP) in the spinal cord was detected by immunohistochemical technique. The Area and the Gray Scale of astrocytes were measured in the dorsal horn by using Image-Proplus. Results The pain threshold (18.41±6.25) in the DM group was lower than (37.40±7.35) in the NC group (P<0.01). The Area (1377.37±271.83) and the Gray Scale (8638.21±1861.90) of astrocytes in the DNP group was significantly higher than in the NC group (P<0.01). Conclusion The level of astrocytes in spinal cord of rats in diabetes neuropathic pain is markedly higher. Inhibiting astrocytic activation in the spinal dorsal horn may represent a novel therapeutic strategy for treating DNP.
