syndecan-1在2型糖尿病患者白细胞表面的表达

 【摘要】 目的 检测T2DM患者外周血白细胞表面syndecan-1的表达。 方法 通过流式细胞仪检测外周血白细胞亚群(中性粒细胞、单核细胞、淋巴细胞)上syndecan-1的表达,并分析syndecan-1的表达与多项临床参数的相关性。 结果 将29例无任何并发症的T2DM患者和24名年龄匹配的健康对照者纳入研究。在中性粒细胞,糖尿病组患者(T2DM组)syndecan-1阳性细胞率(10.372 ± 1.767%)较正常对照组(NC组,3.988 ± 0.673%)显著升高(P = 0.002)。在单核细胞,T2DM组syndecan-1阳性细胞率(2.221 ± 0.310%)与NC组(2.975 ± 0.553%)的差异无统计学意义(P = 0.241)。在淋巴细胞,T2DM组syndecan-1阳性细胞率(25.093 ± 3.865%)与NC组(19.520 ± 3.283%)的差异无统计学意义(P = 0.288)。此外,在T2DM组,中性粒细胞syndecan-1的阳性细胞率与BMI呈正相关(r = 0.572, P = 0.004)。 结论 与健康人群相比,T2DM患者外周血中性粒细胞表面syndecan-1的表达增强,且其阳性表达率与BMI呈正相关。

  【Abstract】 Objective To detect the expression of syndecan-1 on leukocytes of patients with type 2 diabetes mellitus. Methods Expression of syndecan-1 was determined on subsets of leukocytes (neutrophil, monocyte and lymphocyte) by flow cytometry, and potential correlations between syndecan-1 and clinical characteristics were analyzed. Results Twenty-nine patients with type 2 diabetes mellitus (without any diabetic complications,T2DM group) and 24 age-matched healthy subjects (NC group) were enrolled. On neutrophils, percentage of syndecan-1-positive cells was prominently higher in T2DM group than that in NC group (10.372 ± 1.767% vs 3.988 ± 0.673%, P = 0.002). On monocytes, the difference of syndecan-1-positive cells between NC group and T2DM group was not significant (2.221 ± 0.310% vs 2.975 ± 0.553%, P = 0.241). On lymphocytes, the difference of syndecan-1-positive cells between NC group and T2DM group was also insignificant (19.520 ± 3.283% vs 25.093 ± 3.865%, P = 0.288). An association between BMI and percentage of syndecan-1-positive neutrophils of patients with type 2 diabetes was detected (r = 0.572, P = 0.004). Conclusions Compared with normal controls, the expression of syndecan-1 on peripheral neutrophils enhances in subjects with type 2 diabetes, and it is associated positively with BMI.
