RANTES基因启动子区-28C/G多态性与2型糖尿病的相关性研究

  【摘要】目的探讨正常T细胞表达和分泌的活性调节蛋白(RANTES)基因启动子区-28C/G多态性与T2DM及其慢性并发症相关危险因素的相关性。方法运用PCR-RFLP技术对178例T2DM患者(T2DM组)和67例正常对照者(NC组)的RANTES基因-28C/G多态性进行检测。结果NC组和T2DM组间基因型和等位基因频率差异无统计学意义(P>0.05);RANTES基因-28G等位基因携带者血浆HDL-C浓度较非G等位基因携带者低且差异有统计学意义(P<0.05); Logistic回归分析: RANTES基因启动子区-28C/G多态性与HDL-C水平无关(P=0.228,OR=1.777),BMI与HDL-C异常呈正相关(P=0.000,OR=2.562)。结论RANTES基因启动子区-28C/G多态性可能不是昆明地区汉族T2DM发生的危险因素,超重和肥胖可能是T2DM患者HDL-C 异常的危险因素。

  【Abstract】ObjectiveTo investigate the associatoin of RANTES (regulated on activation, normal T-cell expressed and secreted) gene promoter -28C/G polymorphism with type 2 diabetes mellitus (T2DM) and risk factors related to chronic complications.MethodsRANTES promoter -28C/G polymorphism was detected by PCR-RFLP in 178 T2DM patients and 67 normal controls (NC).ResultsThere were no statistical differences in genotype and allele frequencies between NC group and T2DM group. The level of HDL-C in patients with RANTES-28G-positive genotype was significantly lower than with -28G-negtive genotype (P<0.05).Logistic regression analysis showed RANTES promoter -28C/G polymorphism was not associated with the level of HDL-C. BMI was positivly correlated with HDL-C abnormality.ConclusionRANTES promoter -28C/G polymorphism may not be a risk factor for T2DM, while, overweight and obesity may be risk factors for HDL-C abnormality of patients with T2DM in Kunming Han people.
