吡格列酮对高脂血症大鼠主动脉脂联素受体mRNA表达的影响

  【摘要】目的观察高脂血症大鼠主动脉脂联素受体1/2(AdipoR1/2)mRNA表达的改变以及吡格列酮干预效果。方法SD大鼠随机分为正常饮食组(NC组)和高脂饮食组(HF组);HF组造模成功后,随机分为高脂组(HL组)和吡格列酮治疗组(Pio组),检测血脂、主动脉病理,以ELISA法检测血清脂联素水平,以实时荧光定量PCR法检测主动脉脂联素受体mRNA的表达。结果高脂饲料喂养12周,HF组TG、TC及LDL-C的水平明显升高(P<0.01)。给药4周后,与HL组比较,Pio组TG、TC明显降低(P<0.01),血清脂联素明显升高(P<0.05);与NC比较,HL组主动脉脂联素受体1/2mRNA表达明显下降(P<0.05);Pio组主动脉脂联素受体1/2mRNA表达明显升高(P<0.05或P<0.01)。结论脂联素受体表达下降可能介入高脂血症血管损伤,吡格列酮具有改善高脂血症血管损伤及抗动脉粥样硬化作用,该作用可能与提高主动脉血管脂联素受体表达有关。

  【Abstract】ObjectiveTo observe the effect of pioglitazone on expression of adiponectin receptor (AdipoR) of the aorta in hyperlipidemia rats.MethodsSD rats were randomly devided into the high-fat diet group(HF) and normal control group(NC). HF group were fed a high-fat diet for 12 weeks,then the level of serum lipid was measured. SD rats in HF group were randomly devided into two groups: hyperlipidemia group(HL)and pioglitazone treatment group(Pio). Serum lipid level was measured in the rats and the aorta was taken for pathologic analysis. Adiponectin(APN) in serum was measured by ELISA. Real time fluorescent quantitation PCR was used to evaluate AdipoR1/2 mRNA expressions of the aorta.ResultsAfter 12 weeks’ feeding, as compared with NC group, rats fed high-fat diet showed significantly higher levels of triglyceride (TG), total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C) (P<0.01). After four weeks’ treatment, as compared with HL group , the level of TC and TG in Pio group was obviously lower (P<0.01), and the serum APN in Pio group was obviously increased (P<0.05). Treatment with pioglitazone enhanced the mRNA expression of both AdipoR1 and AdipoR2(P<0.05 or P<0.01).ConclusionsDecreased expression of AdipoR may involve in vascular impairment in rats with high-fat diet induced hyperlipidemia. Pioglitazone may exerte a significant vascular protective effect in hyperlipidemia rats, which may relate to increased serum APN and expression of AdipoR in vascular wall and contribute to their anti-atherosclerosis effect.
