间充质干细胞联合骨髓细胞输注对糖尿病胰岛移植大鼠细胞因子的影响

  【摘要】目的探讨同种异基因大鼠骨髓间充质干细胞(MSCs)联合骨髓细胞(BMCs)输注对胰岛移植大鼠体内细胞因子水平的影响。方法制备SD大鼠DM模型,Wistar大鼠骨髓传代纯化获得MSCs。经静脉输注供体MSCs 和/或BNCs后7d,糖尿病SD大鼠进行同种异体胰岛移植,观测胰岛移植物存活时间,检测白介素-2(IL-2)、干扰素-γ(IFN-γ)、白介素-4(IL-4)、白介素-10(IL-10)、白介素-6(IL-6)及转化生长因子-β1(TGF-β1)含量。结果预输注高浓度MCSs联合BMCs能显著延长移植胰岛的生存时间,平均为30d。该组大鼠体内IL-2、IFN-γ水平降低, IL-10和TGF-β1水平升高(P<0.05)。结论高浓度同种异基因大鼠MSCs联合BMCs输入体内后,使胰岛移植大鼠血清细胞因子发生显著变化,可能是诱导胰岛移植免疫耐受的原因之一。

  【Abstract】ObjectiveTo investigate the influence of co-transfusion of mesenchymal stem cells and bone marrow cells before islet transplantation on cytokines in diabetic rat.MethodsAnimal model of diabetes mellitus was established. MSCs from Wistar rats were purified through passage culture. The donor mesenchymal stem cells and/or bone marrow cells were injected into the recipient SD rats via veins, and diabetic SD rats received islet transplantation 7 days after the injection. Survival time of the islets grafts was observed, and the level of serum IL-2,IFN-γ, IL-4, IL-10 and TGF-β1 were detected.ResultsThe pre-infusion of MSCs and BMCs significantly prolonged allograft survival time(MST 30 d). The contents of IL-2 and INF-γ were reduced while the contents of IL-10 and TGF-β1 were increased significantly after co-infusion of high concentration of MSCs and BMCs.ConclusionThe contents of cytokines can be changed by co-transfusion of high-concentration MSCs and BMCs, which might be one of the reasons to induce pancreatic islets transplantation tolerance.
