【摘要】 目的 观察AGE对人脐静脉内皮细胞(HUVEC)标志物表达的影响。 方法 在体外培养的HUVEC细胞株中分别加入AGE、牛血清白蛋白(BSA),采用免疫组织化学技术和Western blot检测HUVEC中α-平滑肌肌动蛋白(α-SMA)和血管性血友病因子(vWF)的表达情况。 结果 HUVEC经AGE处理7 d后,AGE组α-SMA表达较BSA组增强[(27.04+0.05)vs(20.12+0.02),P<0.01],vWF表达较BSA组降低[(29.66+1.44)vs(55.28+1.77),P<0.01]。 结论 AGE可体外诱导HUVEC表面标志物发生改变,该转分化机制可能参与糖尿病血管病变的发生发展。

【Abstract】 Objective To investigate the effect of advanced glycation end products (AGE) on the expression of markers in human umbilical vein endothelial cell (HUVEC). Methods HUVECs were respectively incubated with AGE-BSA and BSA, and then the expression of α-smooth muscle actin (α-SMA) and von willebrand factor (vWF) in HUVECs were detected with immunohistochemistry and western blot method. Results In AGE versus BSA group, the expression of α-SMA were increased [(27.04±0.05) vs (20.12±0.02),P<0.01], and the expression of vWF were decreased [(29.66±1.44) vs (55.28±1.77),P<0.01], after the HUVECs were incubated by AGE-BSA for seven days. Conclusion Exposure to AGE can change the expression of surface markers of HUVECs, this suggeste that the transforming of HUVECs may involve in the pathogenesis of diabetic vascular complications.
