腹腔注射分泌胰升血糖素样肽-1重组腺伴随病毒对纠正db/db小鼠高血糖的实验研究小鼠高血糖的影响

  【摘要】 目的 观察腹腔注射分泌表达胰升血糖素样肽-1(GLP-1)的重组腺伴随病毒对db/db小鼠高血糖的纠正作用。 方法 将30只8周龄db/db小鼠随机分为对照组与实验组。测量体重、血糖。实验组腹腔注射分泌表达GLP-1的重组腺伴随病毒,对照组腹腔注射对照病毒。给药后分别于第2、4、6、8、10、12周测量体重、血糖情况。 结果 随观察时间的延长,对照组体重、血糖呈上升趋势,实验组则下降。实验组2周时,体重(38.3±4.4)g,血糖(18.2±3.0)mmol/LlL,与对照组(44.2±4.7)g、(22.6±4.5)mmol/L比较,差异无统计学意义(P>0.05)。第4-~12周时实验组体重和血糖均低于对照组,两组间比较差异有统计学意义(P<0.05)。 结论 通过腹腔注射分泌表达GLP-1的重组腺伴随病毒方式,可纠正db/db小鼠高血糖。

Regulation effect of Secretory secretory glucagon-like peptide 1 Recombinant recombinant Adenoadeno-associated Virus virus on hyperglycemia in db/db Micemice: A experimental study LOU Ming-wu, LIANG Bing, ZHANG Ming-dong.   【Abstract】 Objective To study the regulation effect of the secretory glucagon-like peptide 1(GLP-1) recombinant adeno-associated virus on hyperglycemia in db/db mice with defective leptin receptor. Methods Thirty eight-week old db/db mice were divided into control and experimental groups randomly(n=15, each). After measuring weight and blood glucose in two groups, the secretory GLP-1 recombinant adeno-associated virus(1×107 pfu, qd) were intraperitoneally injected into experimental group for twice. And the control group were intraperitoneally injected with control virus (1×107 pfu, qd). Weight and blood glucose were measured in two groups at 2, 4, 6, 8, 10, 12 weeks after the final intraperitoneal injection. Results Two weeks after injection, weight and blood glucose were all increased in control group, but decreased in experimental group along with time going, there was no obviously differences in weight and blood glucose between two groups(P>0.05). But the difference in the indexes between two groups was obvious at time points of 4, 6, 8, 10, 12 weeks. Conclusion The intraperitoneal injection of the secretory GLP-1 recombinant adeno-associated virus has regulation effect on high blood glucose in db/db mice.
