糖尿病大鼠心肌组织内脏脂肪素的表达及二甲双胍干预作用的观察

  【摘要】目的 探讨糖尿病大鼠心肌组织内脏脂肪素(Visfatin)、磷脂酰肌醇酶-3激酶(PI3K)表达及二甲双胍对其干预作用。方法 60只SD大鼠随机分为正常对照(NC)组、糖尿病(DM)组、二甲双胍治疗(M+DM)组,成模后测定FBG、FIns、FFA、TG、TC、LDL-C,计算胰岛素抵抗指数(HOMA-IR)、ISI;测定心肌Visfatin、PI3K mRNA表达;测定心肌Visfatin蛋白表达;观察心肌病理变化。结果 DM组FBG、FFA、TG、TC、FIns、HOMA-IR较NC组增高(P<0.05),M+DM组较DM组降低(P<0.05)。DM组心肌组织Visfatin、PI3K mRNA表达较NC组降低(P<0.05),M+DM组心肌组织Visfatin、PI3K mRNA表达均较DM组增高(P<0.05);M+DM组心肌组织Visfatin蛋白表达较DM组增高(P<0.05)。结论 二甲双胍可提高DM大鼠心肌组织Visfatin、PI3K表达,对心肌组织保护的作用机制可能是通过调节心肌Visfatin表达水平进而通过PI3K途径完成。

  【Abstract】 Objective To observe the expressions of visfatin and phosphoinositide 3-kinase (PI3K) in the myocardium of diabetic rats and the changes of their levels induced by the intervention with metformin. Methods Sixty male Sprague-Dawley rats were randomized into the normal control group (NC), diabetes group (DM) and metformin treatment group (M+DM). The levels of FBG, FFA, TG, TC, LDL-C, and FIns of successfully made diabetic models were measured to calculate HOMA-IR and ISI, the expressions of visfatin and PI3K mRNA in the myocardium were determined, the expression of visfatin in the myocardium was detected, and the pathological changes in the myocardium of rats were observed. Results In the DM group, the levels of FBG, FFA, TG, TC, FIns, and HOMA-IR were significantly higher than those in the NC group (P<0.05), in comparing with the DM group, those levels in the M+DM group were decreased (P<0.05). The expressions of visfatin and PI3K mRNA in the myocardium of the DM group was lower than those of the NC group (P<0.05); those in the M+DM group were higher than in the DM group (P<0.05); and the expression of visfatin in the M+DM group was higher than in the DM group (P<0.05). Conclusions Metformin functions to increase the expressions of visfatin and PI3K in the myocardium of DM rats. Its protective functions for the myocardium are probably realized through regulating the expression of visfatin in the myocardium via the PI3K pathway.
