  四氧嘧啶诱导的兔1型糖尿病模型心房重构的实验研究

  【摘要】 目的 研究四氧嘧啶诱导的兔T1DM模型的心房病理和电生理学改变及与炎症氧化应激反应的关系。方法 DM组成年健康长耳白兔通过耳缘静脉注射四氧嘧啶诱导兔T1DM模型。4周后动物麻醉开胸暴露心脏,建立Langendorff灌流的离体兔房颤模型及进行心脏电生理指标的检测。取心房组织制作切片进行HE及Masson染色,测量胶原容积分数。取血检测血清中炎症和氧化应激相关指标。结果 病理学检查显示DM组兔心房显著纤维化(P<0.01)。房间传导时间(IACT),不同S1S1起搏周长时心房有效不应期离散度延长,不同S1S1起搏周长时低位左房有效不应期明显缩短 (P<0.05),其他各点有缩短的趋势,但差异无统计学意义。DM组有6例诱发房颤(P<0.01),Con组未能诱发房颤。DM组血清生化检测显示胰岛素水平下降(P<0.05);葡萄糖、高敏C反应蛋白(hsC-RP)、TNF-α、氧自由基(O2-)水平增高(P<0.05)。结论 四氧嘧啶诱导的兔T1DM伴随着心房病理及电生理的改变,可能与机体的炎症氧化应激反应有关。

Experimental study on atrial remodelling in alloxan induced rabbit T1DM Model ZHAO Hui, LI Guang-ping, LIU Tong. Department of Cardiology, Tianjin Tianhe Hospital, Tianjin 300050, China

  【Abstract】 Objective To study the pathological and electrophysiological changes, and their relationship with oxidative stress and inflammatory response in alloxan induced T1DM rabbit models. Methods Healthy adult rabbits in diabetic group were injected through the marginal ear vein with alloxan to induce rabbit T1DM. After 4 weeks, rabbits were then anesthetized, thoracotomy was performed and pericardium opened to expose heart, the Langendorff perfused isolated rabbit model was established and cardiac electrophysiological parameters were measured. Sections of the atrium tissues were made and with Masson staining, and Collagen volume fraction (CVF) was measured. Inflammatory and oxidative stress parameters were measured. Results The pathological examination showed that a significant atrial fibrosis was observed in the diabetic rabbits (P<0.01). Intra-atrial conduction time (IACT) and AERPD were increased in diabetic group compared with controls, and AERP in lower left atria was shorter in diabetic group (P<0.05). There was a tendency of becoming shorter in other points but the difference was not significant. AF was induced in 6 rabbits in the diabetic group, whereas no AF could be induced in controls (P<0.01). The concentration of insulin was lower (P<0.05), but glucose, hsC-RP, TNF-α, and O2- were higher in the diabetic group than in the control group (P<0.05). Conclusion Alloxan-induced rabbit T1DM is accompanied with atrial pathological and electrophysiological changes, and it may relate to oxidative stress and inflammatory response.
