常规降糖与联合N-乙酰半胱氨酸对糖尿病慢性肾脏脏疾病患者PARP-1及GAPDH活性影响的研究

【摘要】 目的 观察采用常规降糖治疗与联合N-乙酰半胱氨酸(NAC)治疗对糖尿病性肾脏疾病(DN)患者多聚ADP核糖聚合酶(PARP)、3-磷酸甘油脱氢酶(GAPDH)活性的影响。 方法 将60例Ⅲ期DN患者,随机分为常规治疗组(A组)30例;联合NAC治疗组(B组)30例;选取同期健康人群30名为对照(NC)组,治疗12周。观察治疗前后研究对象外周单核细胞PARP活性、全血GAPDH活性的变化。 结果 A、B组治疗后PARP活性下降[A组(3.70±0.06)vs(3.02±0.07)U/mg ;B组(3.74±0.11)vs(2.66±0.04) U/mg],GAPDH活性升高[A组(4430.8±279.4)vs(5136.6±321.6)U/ml,B组(4495.2±324.7)vs(6104.4±275.3)U/ml],且B组治疗后PARP及GAPDH活性改变与A组比较,差异有统计学意义(P<0.05)。 结论 联合NAC治疗与常规治疗比较,可更有效地逆转DN患者PARP、GAPDH活性的异常。

【Abstract】Objectives To observe the effects of conventional treatment and N-Acetylcysteine (NAC) on the activities of PARP and GAPDH in the patients with diabetic nephropathy. Methods A total of 60 patients with DN (stage Ⅲ) were included and randomized into two groups. The conventional group (30 cases) was treated with conventional treatment, and the NAC group (30 cases) was treated with NAC plus conventional treatment. Another 30 healthy people were recruited as controls. PARP activity in peripheral blood mononuclear cells, GAPDH activity in whole blood before and after 12 weeks’ treatment were measured. Results 1. After treatment, the PARP activity in both groups was decreased and GAPDH activity increased (P<0.05) [conventional group (4430.8±279.4) vs (5136.6±321.6)U/ml,NAC group (4495.2±324.7) vs (6104.4±275.3)U/ml]. 2. Changes of PARP and GAPDH activities after treatment in NAC group were more significant (P<0.05). Conclusion Compared with the conventional treatment, the NAC plus conventional treatment can reverse disorders of PARP and GAPDH activities in the DN patients more effectively.
