【摘 要】 目的 探讨胰岛素对血管内皮细胞中miRNA-21的表达和分泌影响。方法 用不同浓度胰岛素(0 nM、1 nM、10 nM、50 nM、100 nM和1000 nM)干预人脐静脉内皮细胞(Human Umbilical Vein Endothelial Cells , HUVECs),48小时后收集细胞上清液及细胞, 用实时荧光定量PCR方法检测细胞及上清液中miRNA-21的表达情况。 结果 与0nM胰岛素组相比,10nM、50nM、100nM和1000 nM胰岛素组HUVECs中miRNA-21表达均下调(p <0.05),1000 nM胰岛素组细胞中miRNA-21表达最低 (p<0.05),但1nM与0nM胰岛素组HUVECs中miRNA-21表达无差异(p>0.05)。与0nM胰岛素组相比,1nM和10 nM胰岛素组HUVECs上清液中miRNA-21表达升高 (p <0.05),而50nM、100nM和1000 nM胰岛素组上清液中miRNA-21表达降低(p <0.05),以50 nM胰岛素组上清液中miRNA-21表达含量最低(p<0.05)。结论 不同浓度胰岛素可影响HUVECs中miRNA-21表达及分泌,这种变化可能共同反映了不同浓度胰岛素对HUVECs的凋亡和增殖影响。

【Abstract】 Objective To explore microRNA-21(miRNA-21) expression and secretion in vascular endothelial cells treated with insulin. Methods Human umbilical vein endothelial cells(HUVECs) were treated with different concentrations of insulin (0nM,1nM,10nM,50nM,100nM and 1000nM respectively) for 48 hours, and then HUVECs and supernate accordingly were collected to detect miRNA-21 expression with Real-time PCR method. Results To compare with the control group of 0nM insulin, miRNA-21 expression were down-regulated in HUVECs with 10nM, 50nM, 100nM and 1000nM insulin groups respectively (p<0.05) and miRNA-21 expression was reduced most significantly in 1000nM insulin group (p<0.05), but there were no difference in miRNA-21 expression in HUVECs between 0nM and 1nM insulin group(p>0.05). To compare with 0nM insulin group, miRNA-21 expressions were up-regulated significantly in cells supernate with 1 nM and 10nM insulin groups (p <0.05), however the expression of miRNA-21 expressions were down-regulated in supernate of 50nM, 100nM and 1000nM insulin groups respectively (p<0.05). Conclusions Different concentrations of insulin could effect the expression and secretion of miRNA-21 in HUVECs , which possibly reflect the apoptosis and proliferation in HUVECs treated with different concentrations of insulin together.
