肾上腺髓质素对高糖培养的肾小球系膜细胞增殖和凋亡的影响及其机制的研究

  【摘要】 目的 观察肾上腺髓质素(AM)对高糖培养的肾小球系膜细胞(HBZY-1)增殖和凋亡的影响及其作用的可能机制。 方法 采用MTT法和流式细胞仪测定观察AM干预对HBZY-1细胞增殖和凋亡的影响,同时采用Western blot观察分裂素激活蛋白激酶 (MAPK)信号传导途径中ERK和P38MAPK蛋白活性的变化。 结果 高糖状态下HBZY-1细胞增殖明显增强,凋亡轻度增加;AM干预后HBZY-1细胞增殖明显抑制,平均下降38.8%,同时凋亡明显增强。AM干预后,ERK1/2活性明显抑制,与高糖组比较平均下降约30%,而P38MAPK活性明显增强,较高糖组平均增高约70%。 结论 AM对高糖培养HBZY-1细胞增殖增强具有抑制作用,并促进其凋亡。这种作用是可能是通过MAPK信号传导途径所介导的。

  【Abstract】 Objective To observe the effects of adrenomedullin (AM) on the proliferation and apoptosis of mesangial cells HBZY-1 cultured under high glucose conditions and further investigate its possible mechanism. Methods The proliferation and apoptosis of mesangial cells HBZY-1 were examined with MTT assay and flow cytometer analysis respectively. The changes of ERK and P38MAPK protein activities in the mitogen activated protein kinase (MAPK) signaling pathway were observed with Western blot assay. Results The proliferation of mesangial cells HBZY-1 was significantly enhanced and the apoptosis was increased under high glucose conditions. After the intervention with different concentrations of AM, the proliferation of HBZY-1 cells decreased by 38.8 percent, while the apoptosis of HBZY-1 cells increased about 1.7 times in the AM (10-8mol/L) group, and 3 times in the AM (10-7mol/L) group compared with the high glucose group. Meanwhile, the activities of extracellular-signal regulated kinase 1/2 (ERK 1/2) protein were significantly decreased by 30 percent and the activities of P38MAPK protein increased by 70 percent in the AM group compared with the high glucose group. Conclusion AM could significantly inhibit the proliferation of mesangial cells and promote their apoptosis under high glucose conditions through the activities of MAPK signal pathway, suggesting that AM might have renal protective properties in diabetic nephropathy.
