【摘要】 目的 评价葡萄糖激酶调节蛋白(GCKR)基因rs780094与中国汉族人群T2DM及其相关性状的关系。 方法 采用病例对照研究,在SEQUINOME技术平台上,对1167名血糖正常者(NC组)和1457例T2DM患者(T2DM组)的GCKR基因rs780094位点进行基因分型,以该位点次要等位基因G为自变量进行Logistic回归分析,并调整协变量性别、年龄和BMI。利用Revman4.2,通过Meta分析系统评价该位点与中国汉族T2DM的相关性。 结果 未发现rs780094位点与T2DM相关,但在显性模式下可见相关趋势(OR=1.23, 95%CI0.99~1.15,P=0.07)。在NC组发现GG基因型个体的FPG、OGTT 0.5和2 h的Ins高于AA基因个体,P值分别为0.04、0.02和0.03。对已发表的中国汉族人群的T2DM与rs780094位点的关联研究进行Meta分析,显示rs780094与中国汉族T2DM相关,OR为1.12,95%CI为1.08~1.17,P<0.00001。 结论 在中国汉族人群中,GCKR基因的rs780094与FPG、胰岛素素敏感性以及T2DM相关。

【Abstract】 Objective To evaluate the association between rs780094 in glucokinase regulatory protein (GCKR) gene and type 2 diabetes and its related traits. Methods A case control study including 1167 subjects with normal oral glucose tolerance(OGTT) and 1457 patients with type 2 diabetes was conducted. All the subjects were genotyped for rs780094 by SEQUINOME and Logistic regression was used to calculate odd ratio(OR) with adjustment of age,sex and body mass index as covariates .By Revman4.2, Meta-analysis was performed to estimate overall OR including all the published studies in Chinese Han. Results No association of rs780094 with type 2 diabetes in our study was replicated, although a tendency toward association was observed under dominant genetic model(OR=1.23, 95% CI, 0.99~1.15,P=0.07)).In control subjects, the subjects with GG genotype had higher fasting plasma glucose,30 min serum insulin and 2 hour serum insulin levels during OGTT as compared with those with AA genotype(P=0.04,0.02 and P=0.03,respectively).Meta-analysis showed the association of rs780094 with type 2 diabetes(OR=1.12,95%CI 1.08~1.17,P<0.00001). Conclusion This study confirmed the association of rs780094 with fasting plasma glucose, type 2 diabetes and insulin sensitivity in Chinese Han.
