甘丙肽对鼠脂肪细胞葡萄糖转运蛋白4膜位移的影响

  【摘要】 目的 探查甘丙肽能否促进大鼠脂肪细胞GluT4膜位移。方法 用3H葡萄糖测3T3 L1脂肪细胞葡萄糖摄取率;大鼠分四组:安静和运动对照组注射生理盐水,安静和运动用药组注射甘丙肽拮抗剂M35。两运动组大鼠游泳20d,60 min/d。 结果 甘丙肽增加葡萄糖摄取;两M35组大鼠正糖钳的葡萄糖输注速率及脂肪细胞GluT4 mRNA水平均下降,细胞外膜和总膜GluT4浓度降低,且外膜与总膜GluT4比值低于对照。 结论 甘丙肽可促进GluT4膜转位而增强胰岛素敏感性。

  【Abstract】Objective To explore the effect of galanin on translocation of glucose transporter 4 (GluT4) in adipocytes of rats. Methods The glucose uptake rate was measured in 3T3 L1 adipocytes with 2-deoxy-[3H]-D-glucose. Rats were randomized into 4 groups: the sedentary and trained control groups were injected saline, and the sedentary and trained treatment groups were injected M35, a galanin antagonist. Rats in both exercise groups swam for 20d, 60min/d. Results Galanin treatment increased the glucose uptake rate, while M35 administration reduced glucose infusing velocity in euglycemic-hyperinsulinemic clamp test, GluT4 mRNA expression and GluT4 concentration. The ratio of GluT4 contents in plasma membranes to total cell membranes in the treatment groups was lower than that in the control groups. Conclusion Galanin may benefit GluT4 translocation to plasma membranes to enhance insulin sensitivity in adipocytes.
