【摘要】 目的 探讨糖调节蛋白(GRP78)在肥胖及肥胖相关疾病中的作用。方法 将30例40~60岁女性患者分为3组:正常对照组(NC组，BMI<23 kg/m2),超重组(OW组，BMI<28 kg/m2)和肥胖组(OB组，BMI>28 kg/m), 每组10例。在其腹部择期手术时取其网膜脂肪组织, 应用RT-PCR和Western-blot检测上述三组患者脂肪组织中GRP78的表达水平。 结果 OB组GRP78 mRNA与蛋白水平(GRP78/β-actin)显著高于对照组(P< 0.05)。GRP78/β-actin与BMI呈正相关( r≥0.75, P<0.05)。 结论 GRP78表达量与BMI呈正相关(GRP78蛋白及mRNA水平OB组>OW组>NC组)。GRP78可作为预测肥胖及T2DM和心血管疾病危险因素的指标之一。

　　【Abstract】 Objectives To investigate the effect of GRP78 in the pathogenesis of obese and obesity-related diseases. Methods 30 female patients were divided into three groups: normal control group(NC,BMI < 23 kg/m2), overweight (OW,BMI<28 kg/m2) and obesity group (OB ,BMI>28 kg/m2),10 persons in each group, age between 40 and 60 years old .Take omental fat from 30 patients undergoing elective surgery. Western blot and RT-PCR were applied to monitor the expression difference of GRP78 ，and the correlation between the expression difference and BMI in omental fat in three groups was studied. Results The expression of GRP78 in omental fat from obesity group were higher than normal control group(p<0.05) ,the expression of GRP78 vs β-actin were positively correlated with BMI in three groups. Conclusions The expression of GRP78 in omental fat are significantly correlated with BMI,GRP78 is a better predictor of risk factors in newly diagnosed type 2 diabetes and cardiovascular disease.
