【摘要】 目的 探讨男性糖尿病(DM)患者血管病变与外周血内皮祖细胞(EPCs)数量及功能的关系。 方法 对15名健康者(NC组)、20例DM未合并血管病变者(DM组)、18例DM合并血管病变者(DVP组)及10例非糖尿病冠心病者(NDCHD组)采集空腹静脉血,分离并培养EPCs。应用流式细胞技术、MTT法、Boyden小室法、黏附法对EPCs进行鉴定、计数,以及细胞增殖、迁移及黏附功能测定。 结果 (1)EPCs与HbA1c呈负相关(r=-0.54,P=0.02)。(2)EPCs数量和增殖功能在NDCHD组< DVP组

The correlation between endothelial progentior cells of peripheral blood and vascular complications in male patients with diabetes mellitus

【Abstract】 Objective To investigate the relationship between the vascular complications and the endothelial progenitor cells(EPCs) of peripheral blood in male patients with diabetes mellitus(DM). Method sThe study individuals included DM group (without vascular complications, n=20) , DVP group (with vascular complications, n=18), normal control (NC, n=15) and CHD control (with coronary heart disease without DM, n=10). The fasting vein blood was collected and EPCs were separated and cultured.Flow cytometry (FCM) was used for identifying surface markers of EPCs, MTT assay for EPCs proliferation, Boyden chamber for evaluating migration of EPCs, and adhesion test for determinating adhering capacity of EPCs. Result s①The number of EPCs had a negative correlation with HbA1c(r=-0.56,P=0.02) and SBP (r=-0.41,P=0.03). ②The numbers of EPCs were 2.97%±1.72%, 4.09%±1.15%, 10.09%±3.21%, 14.76%±5.01% in groups of CHD control, DVP, DM and NC, respectively(P<0.05). ③The absorptivity values of MTT assay for EPCs proliferation were 0.06±0.03, 0.12±0.09, 0.18±0.08, 0.23±0.03 in groups of CHD control, NC, DM and DVP,respectively (P<0.05). ④The migration rate of EPCs was 10.01±3.29, 11.39±3.08, 13.35±3.41 and 23.00±3.01 in groups of CHD control, DVP, DM and NC with no significant difference (P>0.05) between CHD control and DVP group, but it was lower in groups of DM, DVP and CHD control than in NC (all P<0.05). ⑤The adhering capacity had no statistical difference among all four groups(all P>0.05). Conclusion sMale diabetic patients with vascular complication have a decreased number of EPCs and a decreased capacities of EPCs proliferation and migration function, which is positively correlated with vascular complications.
