替米沙坦对糖尿病大鼠胰岛硫氧还蛋白与硫氧还蛋白相互作用蛋白表达的影响

【摘要】 目的 探讨替米沙坦对糖尿病大鼠胰岛硫氧还蛋白(TRX)与硫氧还蛋白相互作用蛋白(TXNIP)表达的影响。 方法 SD大鼠分为正常对照(NC)、糖尿病(DM)和替米沙坦干预(TM)组。DM组和TM组大鼠高脂高糖喂养4周后,腹腔注射链脲佐菌素(STZ)(30mg/kg)构建2型糖尿病(T2DM)模型,TM组予替米沙坦10mg·kg-1·d-1干预,8周后测定血浆生化指标、胰岛中胰岛素、TRX和TXNIP的表达。 结果 DM组和TM组空腹血糖、丙二醛以及胰岛中TXNIP水平均较NC组高(P<0.05),TM组血糖与DM组差异无统计学意义,但TM组上述其余指标水平较DM组低(P<0.05);DM组和TM组空腹胰岛素、过氧化氢酶、超氧化物歧化酶及胰岛中胰岛素和TRX水平低于NC组(P<0.05),但TM组高于DM组(P<0.05)。 结论 T2DM大鼠胰岛TRX表达较正常大鼠降低,而TXNIP升高;氧化应激水平和低胰岛素血症的改善可能与替米沙坦干预了这种变化有关。

Effects of telmisartan on thioredoxin and thioredoxin-interacting protein expression in pancreatic islets of diabetic rats

【Abstract】 Objective To assess the effects of telmisartan on thioredoxin (TRX) and thioredoxin-interacting protein (TXNIP) expression in pancreatic islets of diabetic rats. Methods SD rats were randomly assigned to normal control (N),diabetes (D) and telmisartan (T) groups.Experimental SD rat model with type 2 diabetes mellitus (T2DM) was induced by a combination of 4 weeks’ diet rich in fat and sugar and a single intraperitoneal injection of streptozotozin (STZ) (30mg/kg).Rats in T group were treated with telmisartan 10mg/ (kg · d) for 8 weeks.Plasma biochemical parameters and the expression of insulin,TRX and TXNIP in islets were measured. Results The fasting plasma glucose and malonaldehyde (MDA) level and TXNIP expression in islets were higher in D and T groups than in N group (P<0.05),the differences in levels of fasting plasma glucose between D group and T group were not statistically significant,but the fasting plasma MDA level and TXNIP expression in islets were lower in T group than in D group (P<0.05).The levels of fasting plasma insulin,superoxide dismutase (SOD),catalase (CAT) and expressions of insulin and TRX in islets were lower in D and T groups than in N group (P<0.05),but were higher in T group than in D group (P<0.05). The expression of TRX in islets is lower in T2DM rats than in normal control rats,but expression of TXNIP is higher.Treatment with telmisartan might down-regulate the expression of TXNIP and up-regulate the expression of TRX,which might help to improve the oxidative stress and hypoinsulinemia in T2DM rats.
