糖尿病大鼠胰腺Wnt/β-catenin信号通路的变化及其调节胰岛干细胞分化与增生的意义

  【摘要】 目的 研究胰岛β细胞损伤后胰岛再生过程中Wnt/β-catenin信号通路的生物调控。方法 STZ破坏新生大鼠胰岛β细胞,免疫组织化学观察1 d、2 d、3 d 中Wnt/β-catenin通路β-catenin为核心的上、下游基因APC、c-Myc以及Nestin在胰岛再生过程中的表达。 结果 β-catenin在残留的胰岛细胞中表达上调,并在胰腺间质中异常表达;APC表达于胰腺导管上皮及残留的胰岛细胞;c-Myc则主要表达于胰腺腺泡上皮及胰腺间质内;Nestin表达分布于残留的胰岛细胞和胰腺导管上皮。 结论 Wnt/β-catenin信号通路在胰岛再生中可能通过促进胰腺干细胞分化与增殖而调控。

  The change of Wnt/β-catenin pathway in diabetic rat pancrease and its role in regulating the differentiation and proliferation of pancreatic stem cells

  【Abstract】 Objective To investigate the regulation role of Wnt/β-catenin signal pathway in regulating pancreatic islets regeneration after β-cells injury.MethodSTZ were used to injure β-cells and imunohistochemistry method was adopted to check the expression of several vital factors e.g. APC, β-catenin, c-Myc for Wnt/β-catenin signal pathway and of nestin in pancreatic islets regeneration. Results Immunohistochemistry showed that the β-catenin was up-regulated in β-cells and abnormally expressed in pancreatic stroma; APC was expressed on duct epithelium and β cells; C-myc was expressed in stroma and gland alveolus epithelium; nestin was expressed on islet cell and duct epithelium.ConclusionThe regulation roles of Wnt/β-catenin signal pathway in pancreatic islets regeneration may be through promoting pancreatic stem cells differentiation and proliferation.
