【摘要】目的 探讨脂联素对高浓度葡萄糖诱导的原代大鼠胰岛β细胞凋亡的影响及其可能机制。方法 分离和培养原代胰岛细胞,分组干预并检测胰岛细胞凋亡率及相关基因表达水平。结果 高糖组胰岛β细胞凋亡率、Caspase-3和Bax mRNA表达量显著高于正常葡萄糖和高糖联合脂联素组(P<0.01)。结论 脂联素可以抑制高糖诱导的大鼠胰岛β细胞凋亡,其作用机制可能与凋亡相关基因Caspase-3与Bax基因表达下调有关。

【Abstract】Objective To explore the effect of adiponectin on high glucose-induced rat primary pancreatic β-cell apoptosis and the possible mechanism. Method Rat primary islet cells were isolated and divided into several groups, and β-cell apoptosis and mRNA expressions of some apoptosis-related genes were studied. Result The β-cell apoptotic rate and expressions of caspase-3 and Bax mRNA were significantly higher in HG group than in NG and HGAPN (HG plus adiponectin) group. Conclusion Adiponectin can inhibit high glucose-induced primary rat pancreatic β-cell apoptosis and its mechanism might be related to the down regulation of Caspase-3 and Bax mRNA expression.
