【摘要】 目的 观察DN大鼠肾脏解偶联蛋白2(UCP2)的表达并探讨促红细胞生成素(EPO)对UCP2表达的影响。 方法 STZ构建糖尿病大鼠模型,随机分为糖尿病(DM)组,促红细胞生成素干预(EPO)组,另设健康对照(NC)组。PAS和HE染色观察 各组肾脏病理变化,免疫组织化学及Western blot 观察肾组织UCP2蛋白表达水平,RT-PCR测定各组肾组织UCP2 mRNA表达。 结果 DM组4、8、12周肾脏中UCP2mRNA及蛋白的表达逐渐升高,EPO组肾脏UCP2 mRNA及蛋白表达均较DM组减少P<0.05)。 结论 糖尿病大鼠肾脏中UCP2表达升高,EPO可减少UCP2表达,对肾脏起保护作用。

Expression of uncoupling protein 2 in the kidney of diabeticrats and the intervention effect of erythropoietin
【Abstract】 Objective To investigate the expression of uncoupling protein 2(UCP2) in the kidney of diabetic rats and the effect of erythropoietin(EPO)on its expression. Methods Diabetic rat models were established by intraperitoneal injection with STZ.The diabetic models were randomly divided into two groups: diabetic group (DM) and EPO treated group (EPO). Healthy rats were enrolled as control group (NC). The patho-logical changes of the kidney were evaluated by PAS and HE staining.The expression of UCP2 in renal tissue were measured by immunohistochemistry and Western-blot, and the experssion of UCP2 mRNAwas detected by Real-time PCR. Results The mRNA level ofUCP2 was gradually increased from week 4,8,and 12. The mRNA and protein ex-pression ofUCP2were significantly lower in EPO group than in DM group(P<0.05). Conclusion The expression of UCP2 is increased in the kidney of diabetic rats. EPO can protect diabetic kidney by decreasing the expression of UCP2.
