　　【摘要】目的 观察糖尿病大鼠胃窦平滑肌自噬相关蛋白Beclin-1和LC3的变化，探讨其在糖尿病胃动力障碍发生过程中的意义。 方法 将大鼠分为模型2周(DM2w)、模型4周(DM4w)和正常对照(NC)组，检测各组胃窦平滑肌Beclin-1和LC3蛋白表达。 结果 与NC组相比，DM2w、DM4w组胃窦平滑肌Beclin-1蛋白相对表达量均较高[(0.13±0.04)vs(0.22±0.12)，P<0.05;(0.13±0.04)vs(0.62±0.07)，P<0.01]，且DM4w组高于DM2w组[(0.62±0.07)vs(0.22±0.12)，P<0.01]。LC3蛋白显示LC3-Ⅰ和LC3-Ⅱ两条带，与NC组相比，DM2w、DM4w组胃窦平滑肌LC3-Ⅱ/LC3-Ⅰ比值均较高[(0.98±0.09)vs(2.28±0.20)，P<0.01;(0.98±0.09)vs(2.34±0.21)，P<0.01]，DM2w组与DM4w组比较差异无统计学意义[(2.28±0.20)vs(2.34±0.21)，P>0.05]。 结论 自噬相关蛋白Beclin-1和LC3的表达在糖尿病胃动力障碍发病前随病程逐渐加强，自噬可能影响糖尿病胃动力障碍发生。

　　【Abstract】 Objective To study STZ-induced diabetic rats antral smooth muscle Beclin-1 and LC3 chang of expression, and explore the role and significance of them in the occurrence of diabetic gastroparesis. Methods According to Preliminary results of after four weeks rats STZ-induced occurs diabetic gastroparesis. Wistar rats were divided into model two weeks (DM2w) group, model four weeks (DM4w) group and normal control (NC) group. Western blotting was used to detect the antral smooth muscle Beclin-1 and LC3 expression. Results Beclin-1 expression in the value of NC group was (0.13±0.04), DM2w group was (0.22±0.12), DM4w group was (0.62±0.07). There was statistically significant difference between DM4w group and NC group (P<0.01), DM2w and DM4w group (P<0.01), DM2w group and NC group (P<0.05). LC3 protein showed LC3-Ⅰ and LC3-Ⅱ two bands, and LC3-Ⅱ/LC3-Ⅰ ratio of NC group was (0.98±0.09), DM2w group was (2.28±0.20), DM4w group was (2.34±0.21). There was statistically significant difference between DM4w group and NC group (P<0.01), DM2w group and NC group (P<0.01). Conclusion The Beclin-1 and LC3 of expression were gradually strengthened with the disease befor occuring diabetic gastroparesis, and autophagy may affect the incidence of diabetic gastroparesis.
