【摘要】 目的 探讨IGT者及T2DM患者尿8-羟基脱氧鸟苷(8-OHdG)的水平。 方法 选取IGT者(IGT组)、T2DM患者(T2DM组)各40例,另选取正常对照(NC)者40名。收集24 h尿,通过ELISA定量检测尿液中8-OHdG含量,苦味酸法测定尿肌酐(Ucr)浓度,采用酶法测定血糖、肾功能,高效液相法测定HbA1c,Clauss法测定纤维蛋白原(FIB)。 结果 8-OHdG在NC、IGT、T2DM组逐次递增[(6.97±2.13) vs (9.32±2.51) vs (12.26±3.57)] ng/mgCr,比较差异有统计学意义(P<0.05)。经相关分析及多元逐步回归分析发现,2 hPG与尿8-OHdG呈正相关,对尿8-OHdG水平影响较大。 结论 (1)IGT和T2DM患者尿8-OHdG水平升高;(2)2 hPG是影响体内氧化应激(OS)的主要因素。

【Abstract】 Objective To investigate the level of urinary 8-Hydroxy-2’-deoxyguanine (8-OHdG) in patients with IGT and type 2 diabetes . Methods 40 patients with type 2 diabetes (T2DM group), 40 IGT patients(IGT group), and 40 normal control(NC group) were selected. We collected their 24 hours urine, then tested urinary 8-OHdG by enzyme linked immunosorbent assay (ELISA), and tested urine creatinine by trinitrophenol method. Blood glucose, creatinine, uric acid and blood urea nitrogen were tested by enzymology method , and HbA1c was tested by HPLC method, and fibrinogen (FIB) were tested by von Clauss method. Results The level of urinary 8-OHdG group was increased in NC,IGT and T2DM gradually[(6.97±2.13) vs (9.32±2.51) vs (12.26±3.57)] ng/mgCr(P<0.05).Stepwise regression analysis showed that 2 hPG was positively related to the level of urinary 8-OHdG, and had more influence of urinary 8-OHdG. Conclusion (1)The urinary 8-OHdG level is increased in IGT and T2DM patients.(2) The main affect factor of the oxidative stress was 2 hPG .
