国产门冬胰岛素注射液在正常动物中的降血糖作用的评价

  【摘要】 目的 评价国产门冬胰岛素注射液的降血糖作用 方法 正常ICR小鼠和Wistar大鼠被分为5组,分别注射国产门冬胰岛素注射液0.3、0.6、1.2 U/kg和诺和锐0.6 U/kg,观察其对动物FPG和OGTT后血糖变化的影响,并与进口药品诺和锐进行比较。 结果 国产门冬胰岛素注射液在0.3~1.2 U/kg剂量范围内能够明显降低ICR小鼠和Wistar大鼠的FPG以及OGTT后的血糖和血糖曲线下面积(P<0.05),量效关系良好,10~20 min内起效,40~60 min作用达峰,对小鼠和大鼠FPG最大抑制百分数分别为56.6%和47.8%,作用强度和特点与诺和锐相似。 结论 国产门冬胰岛素注射液具有显著的降血糖作用,与进口药品诺和锐相当。

  【Abstract】 Objective To evalutate the hypoglycemic effects of domestic insulin aspart. Methods Normal ICR mice and Wistar rats were divided into five groups (n =10), three groups of animals were separately given domestic insulin aspart at the doses of 0.3 U/kg, 0.6 U/kg and 1.2 U/kg by subcutaneous injection, and one group animals were given NovoRapid at the dose of 0.6 U/kg. Their effects on fasting blood glucose and blood glucose changes by oral glucose loading (2 g/kg) were observed and compared with NovoRapid. Results The domestic insulin aspart in doses from 0.3 U/kg to 1.2 U/kg could significantly decrease the fasting blood glucose and the area of under blood glucose curve (AUC) after an oral glucose loading with good dose-effect relationship in normal ICR mice and Wistar rats. The effects occurred at 10~20 min and the maximum action was at 40~60 min after injection, with the maximum inhibiton percentage in mice and rats were 56.6% and 47.8%, respectively, which were nearly equivalent to NovoRapid. Conclusions The domestic insulin aspart injection shows significant hypoglycemic effect in normal ICR mice and Wistar rats, and the onset time and intensity of the hypoglycemic effects are equivalent to insulin aspart (Novorapid).
