血管内皮生长因子基因SNPrs3025039多态性与2型糖尿病患者相关性的研究

  【摘要】目的 探讨T2DM患者血管内皮生长因子(VEGF)基因SNPrs3025039多态性。方法 运用PCR-RFLP检测170例T2DM(T2DM组)患者和100名健康体检者(NC组)的VEGF基因SNPrs3025039多态性。结果(1)两组的基因型和等位基因频率差异无统计学依依(P>0.05),但糖尿病性慢性肾脏疾病(CKD)组CT、TT基因型频率和T等位基因频率高于糖尿病CKD(NCKD)组(P<0.05); CC基因型和C等位基因频率低于NCKD组(P<0.05)(2)T2DM组BMI、SBP、DBP、HbA1c、FBG、2 hBG、TG水平高于NC组(P<0.01),HDL-C水平较NC组降低(P<0.01)。结合FBG、TG、TC、LDL-C及HDL-C进行多重Logistic 回归分析显示, CT 和TT 基因型与CKD的发生呈正相关(OR=2.75,P <0.05)。结论 T2DM患者VEGF基因SNPrs3025039多态性与正常人群差异无统计学意义,但CT、TT基因型与CKD的发病风险有关。

  【Abstract】 Objective To explore the polymorphism of vascular endothelial growth factor (VEGF) gene (SNPrs3025039) in the development of T2DM. Methods The polymorphism of VEGF gene (SNPrs3025039) was detected in 170 T2DM patients and 100 controls by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results (1). Genotype and allele frequencies did not show significant differences between the two groups (P>0.05). T2DM Patients with chronic kidney disease (CKD) showed a higher frequency of the CT and TT genotype and T allele (P<0.05) and a lower frequency of the CC genotype and C allele (P<0.05) than those who were without CKD. (2). The levels of BMI, SBP, DBP, HbA1c, FBG, 2hBG and TG of the T2DM group were significantly higher and the level of HDL-C was obviously lower than those of controls (P<0.01). The multiple logistic regression analysis with FBG, TG, TC, LDL-C, and HDL-C in combination showed that CT and TT genotypes were positively correlated with the occurrence and development of CKD (OR=2.75, P<0.05). Conclusion The polymorphism of VEGF gene (SNPrs3025039) is not significantly different from that of the controls, but their CT and TT genotypes are positively correlated with the occurrence and development of CKD.
