  【摘要】 目的 探讨抵抗素对β细胞胰-十二指肠同源盒因子-1(PDX-1) 的表达、胰岛素分泌及细胞增殖、凋亡的影响。 方法 βTc6细胞分为对照(Con)组和抵抗素组(浓度20~200 ng/ml),干预6,12,24 h,CCK-8法测定细胞增殖能力,DNA Ladder检测细胞凋亡,RT-PCR检测PDX-1基因表达,放射免疫法测定胰岛素分泌。 结果 (1)与Con组比较,20、40 ng/ml抵抗素组各干预时段,β细胞的PDX-1表达、增殖活力、凋亡、胰岛素分泌差异均无统计学意义(P>0.05);(2)随抵抗素浓度在60~200 ng/ml范围内增加,干预时间由6~24 h延长,细胞PDX-1表达、增殖活力、胰岛素分泌均呈现浓度和时间依赖性降低(P均<0.05);且细胞凋亡条带逐渐明显。 结论 在一定的浓度和时间范围内,抵抗素可呈浓度和时间依赖性下调β细胞PDX-1表达,损害胰岛素分泌,抑制细胞增殖、诱导其凋亡。

  【Abstract】 Objective To investigate the effects of resistin on the gene expression of pancreatic duodenal homeobox-1 (PDX-1), insulin secretion (IS), cell proliferation, and apoptosis in βTc6 cells. Methods βTc6 cells were divided into control group and resistin group (20, 40, 60, 100, 200 ng/ml respectively), and intervened for 6h, 12h, and 24h. The cell proliferation ability was examined by CCK-8, cell apoptosis rate was detected by DNA laddering assay, the mRNA of PDX-1 was detected by reverse transcription polymerase chain reaction (RT-PCR), and IS was observed in every group by radioimmunoassay. Results (1) Compared with the control group, there was no variation in PDX-1 expression, cell proliferation, apoptosis, and IS in 20~40 ng/ml resistin groups during the intervention time (P>0.05). (2) With the level of resistin increased in the range of 60~200 ng/ml and the intervention time prolonged from 6h to 24h , there existed a time- and concentration-dependent decrease of expression of PDX-1, cell proliferation, and IS in βTc6 cells (P<0.05), and the cell apoptosis was induced gradually. Conclusion In a certain range, resistin showed a time- and concentration-dependent manner of gene expression of PDX-1 and cell proliferation inhibition, cell apoptosis induction, and IS function impairment in β cell.
