【摘要】 目的 研究促性腺激素释放激素(GnRH)激动剂(GnRH agonist, GnRHa)对In-R1-G9细胞内胰升血糖素表达的调控作用。 方法 分别用终浓度为0.1 nmol/L、10 nmol/L、1000 nmol/L的GnRHa处理培养的In-R1-G9细胞2 h(对照组用生理盐水处理),通过Western blot和荧光定量PCR技术观察胰升血糖素在蛋白水平及转录水平的变化。 结果 与对照组相比,低浓度的GnRHa(0.1 nmol/L)使胰升血糖素在蛋白水平和转录水平的表达均有所升高,但差异无统计学意义(P>0.05);而高浓度的GnRHa(10 nmol/L、1000 nmol/L)能明显增强胰升血糖素的表达(P<0.05或P<0.01)。 结论 一定浓度的GnRHa能增强In-R1-G9细胞内胰升血糖素的表达。

【Abstract】 Objective To study the effect of GnRH agonist (GnRHa) on the expression of glucagon in In-R1-G9 cells. Methods GnRHa was added to the media of the cells at final concentrations of 0.1, 10, 1000 nmol/Lol/L, and normal saline added as control. At the end of the incubation, expressions of protein and mRNA of glucagon in In-R1-G9 cells were measured by Western blot and real time fluorescence PCR respectively. Results As compared with control, high concentrations (10 nmol/L and 1000 nmol/L) of the GnRHa showed a significantly increased expressions of glucagon (all P<0.05) while low concentration (0.1 nmol/L) did not show any significant change (all P<0.05). Conclusion GnRHa at 10-1000 nmol/L may increase the expressions of glucagon in In-R1-G9 cells.
