过量醛固酮导致SD大鼠胰岛素抵抗

【摘要】  目的  探讨过量醛固酮对SD大鼠血糖和胰岛素敏感性的影响。  方法  雄性SD大鼠随机分为正常对照组(NC)、醛固酮组(ALD)、醛固酮+安体舒通组(ALD+SPI)。观察4周,每周测量空腹血糖、体重和血压,实验第4周行腹腔葡萄糖耐量和腹腔胰岛素耐量试验,检测血脂联素水平。  结果  醛固酮作用4周,SD大鼠空腹血糖变化无统计学意义(P>0.05);ALD组腹腔葡萄糖耐量试验结果与NC组比较差异无统计学意义(P>0.05);腹腔胰岛素耐量试验显示ALD1530 min血糖下降幅度小于NC组(P<0.05)。ALD组血清脂联素水平较NC组下降22.79%(P<0.05)。  结论  过量醛固酮作用4周可导致SD大鼠胰岛素抵抗。

Excess aldosterone induces insulin resistance in SD rats

Abstract  Objective  To investigate the effect of excessive aldosterone infusion on glucose metabolism and insulin sensitivity in SD rats.   Methods  Male SD rats were randomized into three groups: normal control(Con); aldosterone continuously subcutaneously infusion(0.75 μg/h) for 4 weeks (ALD)  via an osmotic minipump; aldosterone plus spironolactone (100 mg·kg-1·d-1 by gavage) (ALD+SPI) . Fasting blood glucose, blood pressure and body weight were measured every week. At the end of the experiment, intraperitoneal glucose tolerance test and intraperitoneal insulin tolerance test (IITT) were performed. Adiponectin concentration was assayed.   Results  No change occurred in fasting blood glucose during aldosterone infusion(P>0.05). There were no differences in results of intraperitoneal glucose tolerance test among three groups (P>0.05). In contrast, the decreased extents of blood glucose at 15 and 30 min during IITT were lower in ALD group than in control group. The level of serum adiponectin in ALD group reduced by 22.79 as compared with Con(P<0.05).   Conclusion  Excess aldosterone can induce insulin resistance in SD rats.
