【摘要】目的 探讨不同脂肪酸对胰岛β细胞功能及内质网应激(ERS)的影响。 方法 选取40只大鼠分为正常饮食对照(N)组、单不饱和脂肪酸饮食(M)组、多不饱和脂肪酸饮食(P)组和饱和脂肪酸饮食(S)组。干预8周行静脉葡萄糖耐量实验(IVGTT),测定胰腺磷酸化真核生物翻译起始因子2的α亚基(p-eIF2α)和CCAAT/增强子结合蛋白(C/EBP)同源蛋白(CHOP)的表达。 结果 8周末,S组IVGTT胰岛素第1、3、5、8 min均较N组降低(P<0.05)。M、P、S组急性胰岛素反应(AIR)均较N组降低(P<0.05或P<0.01)。P、S组胰腺β细胞内p-eIF2α和CHOP的表达较N组增加(P<0.05或P<0.01)。 结论 不同高脂肪酸饮食均可造成胰岛β细胞功能损伤,可能与ERS有关。

【Abstract】 Objective To investigate the effects of dietary fatty acids on pancreatic beta cell function and endoplasmic reticulum stress (ERS). Methods 40 male wistar rats were randomly divided into four groups, a control (N) group and three groups fed with different high fatty acids diets, including high monounsaturated fatty acids diet (M) group, high polyunsaturated fatty acids diet (P) group and high saturated fatty acids diet (S) group. After eight weeks of feeding, intravenous glucose tolerance test was performed[l1] . α-subunit of eukaryotic translational initiation factor 2 (p-eIF2α) and C/EBP (CCAAT/enhancer-binding protein) homologous protein (CHOP) expressions in pancreatic β cells were tested. Results After eight weeks of diet intervention, serum insulin concentrations at 1, 3, 5, 8 minute of S group decreased significantly compared N group (P<0.05). The AIRs of M, P, S groups were significantly lower than N group (P<0.05 or P<0.01). The protein expressions of p-eIF2α and CHOP in S and P group were significantly higher than N group (P<0.05). Conclusion High fatty acids diets impaired β cells function, which may be associated with ERS.
