microRNAs103和107与胰岛素抵抗的关系

      【摘要】 目的 测定以IR为主的超重或肥胖T2DM患者血清microRNAs103和microRNAs107(miR-103/107)的表达水平,研究miR-103/107与IR的关系。 方法 选取超重或肥胖且IR的新诊断T2DM患者(IR组)50例及健康对照(NC)者30名,采用RT-PCR方法测定血清miR-103/107的表达水平,同时检测FPG、FIns及TG,采用稳态模型评估胰岛素抵抗指数(HOMA-IR)。 结果 (1)IR组血清miR-103/107的表达水平高于NC组[(5.77±0.82/8.84±1.04) vs (2.53±0.51/4.51±0.94),P<0.05];(2)Pearson相关分析显示,miR-103/107分别与HOMA-IR、TG及BMI呈正相关;(3)miR-103/107是IR的独立影响因素(β=0.430、0.054,P=0.016、0.010)。 结论 miR-103/107参与IR的形成,可能作为治疗IR的靶点之一。

  【Abstract】 Objective To determine the expression levels of serum microRNAs103 and 107 (miR-103/107) in type 2 diabetes mellitus with overweight or obesity, to study the relationship between miR-103/107 and insulin resistance. Methods 50 newly diagnosed T2DM patients with overweight or obesity(IR group)and 30 healthy cases (NC group) were collected. The expression levels of miR-103 and 107 were detected by quantitative PCR. FPG, FIns and TG were measured. The basal insulin resistance (HOMA-IR) was evaluated by homeostatic model assessment. Results (1)The expression levels of serum miR-103/107 were higher in IR group than in NC group[(5.77±0.82/8.84±1.04) vs (2.53±0.51/4.51±0.94), P<0.05].(2) Pearson correlation analysis showed that miR-103/107 were respectively positively correlated with HOMA-IR, TG and BMI.(3) Multiple linear regression analysis showed that miR-103/107 were independently influencing factors of HOMA-IR(β=0.430,0.054, P=0.016,0.010). Conclusion miR-103 and 107 involve in the insulin resistance and may be one of target points for the treatment of insulin resistance.
