小檗碱对2型糖尿病地鼠脂肪组织肝X受体通路表达的影响

摘要 目的  观察小檗碱对T2DM地鼠脂肪组织肝X受体(LXR)及其靶基因表达的影响。  方法  以高脂饮食结合小剂量STZ的方法建立IRT2DM地鼠模型。成模后随机分成正常对照(Con)组、IR组、T2DM组、小檗碱治疗(BBR)组,治疗9周。  结果  T2DM组相比,BBRFPG[(6.19±0.84) vs (13.10±0.93) mmol/L]OGTT 2 hPG[(8.83±1.28) vs (15.49±1.31) mmol/L]HOMA-IR[(7.40±1.40) vs (14.80±2.10)]降低(P0.05);RT-PCR结果提示,LXRα[(3.29±0.68) vs (-5.56±-1.23)]LXRβ[(3.09±0.45) vs (-5.49±-0.95)]基因表达上调,并伴随着LXR靶基因表达的改变(P0.05);Western blot结果提示,LXRα[(0.68±0.12) vs (0.11±0.03)]LXRβ[(0.70±0.14) vs (0.13±0.03)]蛋白表达上调(P0.05)。  结论  LXR及其靶基因参与小檗碱治疗T2DM地鼠脂诱性脂肪组织IR的分子机制。

Effect of berberine on liver X receptor expression of adipose tissuein type 2 diabetic hamsters

Abstract  Objective  To investigate the effect of berberine on the expression of adipose tissue liver X receptors (LXRs) and their target genes in type 2 diabetic hamsters.  Methods  The insulin-resistant and type 2 diabetic hamster models were induced by high-fat diet and low-dose streptozotocin.The modeled hamsters were randomly divided into 4 groups: normal control group (Con), insulin-resistant group (IR), type 2 diabetic group (T2DM) and berberine-treated type 2 diabetic group (BBR) for nine-week.  Results  Compared with T2DM group, FPG, OGTT 2 hPG and HOMA-IR in BBR group were significantly lower [(13.10 ± 0.93) vs 6.19 ± 0.84mmol/L; 15.49 ± 1.31vs8.83 ± 1.28mmol/L; 14.80 ±2.10vs 7.40 ± 1.40),P0.05)]. Real-time PCR showed that gene expression of LXRα[(-5.56±-1.23) vs 3.29 ± 0.68] and LXRβ[(-5.49 ±-0.95) vs 3.09 ± 0.45] in BBR group significantly increased with changes of LXR target genes expression (P0.05). Western blot showed that protein expression of LXRα 0.11±0.03vs 0.68±0.12and LXRβ 0.13±0.03vs 0.70±0.14 in BBR group significantly increased (P0.05).  Conclusion  LXR and its target genes are involved in the molecular mechanism of berberine in treatment of adipose tissue insulin resistance in type 2 diabetic hamsters.
